p35/cdk5 binds and phosphorylates beta-catenin and regulates beta-catenin/presenilin-1 interaction

The neuronal cyclin-dependent kinase p35/cdk5 comprises a catalytic subunit (cdk5) and an activator subunit (p35). To identify novel p35/cdk5 substrates, we utilized the yeast two-hybrid system to screen for human p35 binding partners. From one such screen, we identified beta-catenin as an interacting protein.

The APC gene is mutated in the majority of colorectal tumors. The product of this gene is a 300 kDa protein associated with beta-catenin and the human homolog of the Drosophila tumor suppressor DLG. The study of the function of APC provides important insights into the mechanisms of the development of colorectal tumor.

Geldanamycin abrogates ErbB2 association with proteasome-resistant beta-catenin in melanoma cells, increases beta-catenin-E-cadherin association, and decreases beta-catenin-sensitive transcription.

Beta-catenin undergoes both serine and tyrosine phosphorylation. Serine phosphorylation in the amino terminus targets beta-catenin for proteasome degradation, whereas tyrosine phosphorylation in the COOH terminus influences interaction with E-cadherin.
