Hepatic Transport of Bile Acids

The liver is responsible for the secretion of bile acids into bile and for the uptake and reexcretion of bile acids that participate in the enterohepatic circulation. In addition, the secretion of bile acids by the liver is of major importance for the secretion of bile itself and for the secretion of biliary lipids.

Interaction of sulfonylureas with the transport of bile acids into hepatocytes

The sulfonylurea compounds glisoxepide and glibenclamide inhibit the uptake of bile acids into isolated rat hepatocytes. The Ki values for the inhibition of cholate uptake was 9 microM with glibenclamide and 200 microM with glisoxepide. The inhibition of cholate uptake by both sulfonylureas was noncompetitive.

People with genetic or acquired defects in the biosynthesis of bile acids may suffer from cholestasis. Patients with a deficiency of 3β‐hydroxy‐Δ5‐C27‐steroid dehydrogenase/isomerase form 3β,7α‐dihydroxy‐and 3β,7α,12α‐trihydroxy‐5‐cholenoic acids, the sulfated and partially glycine‐conjugated forms of which are found in their urine and bile.

Influence of side-chain charge on hepatic transport of bile acids and bile acid analogues

The importance of side-chain charge on hepatic uptake and biliary secretion of bile acids and analogues was studied using the isolated, perfused rat liver and the anesthetized rat with a bile fistula. Derivatives of cholic acid with negative, neutral, zwitterionic, or positive charges on the side chain were synthesized and studied.
