Micellar electrokinetic chromatography (MEKC) has been exploited as an analytical method for the separation as well as for the determination of serum bilirubin fractions (alpha-, beta-, gamma- and delta-fraction), the activity of bilirubin oxidase (BOD), and for monitoring the catalytic reaction of BOD. The samples were followed by detection at 450 nm.

Purification and properties of bilirubin oxidase fromMyrothecium verrucaria
Applied Biochemistry and Biotechnology, 1991Bilirubin oxidase was purified from a culture filtrate of Myrothecium verrucaria Mv 2, 1089 by DEAE-cellulose and Sephadex G-100 column chromatographies. The purified enzyme had a specific activity of 30 U/mg protein and showed a single band on polyacrylamide gel electrophoresis. Some of the general properties of this bilirubin oxidase were as follows:

A new enzymatic assay for selectively measuring conjugated bilirubin concentration in serum with use of bilirubin oxidase (BOD) has been developed. At pH 5.5 BOD can oxidize only conjugated bilirubin in the presence of reagents such as sodium fluoride and N-acetylcysteine which can decrease BOD reactivity to unconjugated bilirubin and bilirubin ...

The chemical derivatization of bilirubin oxidase (BOx) with a fluorescein derivative (FS) yields a chemically modified enzyme (BOx-FS), with excitation and emission maxima at 487 and 520 nm, respectively. During the oxygen oxidation reaction of bilirubins, in the presence of the modified enzyme, the change in the fluorescence of the modified enzyme ...

AbstractThe unbound bilirubin concentration and the enzymatic rate of bilirubin degradation by bilirubin oxidase in bilirubin‐serum albumin solutions have been investigated experimentally and theoretically. A stoichiometric bilirubin‐serum albumin binding analysis shows that the unbound bilirubin concentration depends only on the molar ratio of the ...

Background We occasionally encounter increases in direct bilirubin value on reanalysis of the surplus serum collected in the past from a neonate with indirect hyperbilirubinemia. But the details of this phenomenon are unclear. We evaluated the change of direct bilirubin and the relation of bilirubin photoisomer of the serum exposed to room light ...
