Clostridium botulinum type D neurotoxin: Purification and detection

Toxicon, 1989
A method is reported for the purification of type D botulinum toxin using a combination of low and high pressure ion exchange chromatography. The procedure produced homogeneous toxin in its free form in 3 days, with a specific toxicity in mice of 5.4 x 10(7) LD50/mg protein.

Modification of carboxyl groups in botulinum neurotoxin types A and E

Toxicon, 1989
Effects of chemical modification of carboxyl groups of botulinum neurotoxin serotypes A and E were studied by using a water soluble carbodiimide-nucleophile reaction that is highly specific for modifying carboxyl groups of proteins. In both types A and E, increasing levels of the reagents, 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide and norleucine ...

Purification and amino acid composition of type A botulinum neurotoxin

Toxicon, 1984
A method to purify type A botulinum neurotoxin from a 64 liter bacterial culture is reported. The procedure includes cation exchange chromatography at pH 7.0. The final product, essentially homogeneous (according to polyacrylamide gel-sodium dodecylsulfate electrophoresis), is a mixture of two forms of the neurotoxin (mol.

Crystal structure of botulinum neurotoxin type A and implications for toxicity

Nature Structural Biology, 1998
Botulinum neurotoxin type A (BoNT/A) is the potent disease agent in botulism, a potential biological weapon and an effective therapeutic drug for involuntary muscle disorders. The crystal structure of the entire 1,285 amino acid di-chain neurotoxin was determined at 3.3 A resolution.

Treatment of Blepharospasm with Botulinum Neurotoxin Type A: Long-Term Results

European Journal of Ophthalmology, 2003
Purpose To describe the long-term efficacy and side effects of treatment of blepharospasm with botulinum neurotoxin type A (Botox). Methods A total of 178 patients with blepharospasm were treated by injections of botulinum toxin in the

Variations in Expression and Release of Botulinum Neurotoxin in Clostridium botulinum Type A Strains

Foodborne Pathogens and Disease, 2007
In this study, we characterized the production and release of botulinum neurotoxin during growth of two Clostridium botulinum strains, Hall A and NCTC2916, of distinct gene organizations. The intra- and extracellular fractions of the bacterial cells harvested at various stages of growth were analyzed for the neurotoxin.

[Effect of urea on botulinum neurotoxin type A].

Molekuliarnaia biologiia, 1983
Kinetic and concentration-mediated dependences of urea effects on toxicity and upon values of negative extremes (279, 287, 292 nm), shifts of lambda max fluorescence from 332 nm to the red region, decrease of fluorescence intensity, changes in accessibility of SH-groups were found.

Characterization of the Genes Encoding the Botulinum Neurotoxin Complex in a Strain of Clostridium botulinum Producing Type B and F Neurotoxins

Current Microbiology, 1998
The organization of the clusters of genes encoding proteins of the botulinum neurotoxin (BoNT) progenitor complex was elucidated in a strain of Clostridium botulinum producing type B and F neurotoxins. With PCR and sequencing strategies, the type B BoNT-gene cluster was found to be composed of genes encoding BoNT/B, nontoxic nonhemagglutinin component (

[Characterization of the subunits of botulinum neurotoxin type A].

Biokhimiia (Moscow, Russia), 1984
A comparative amino acid analysis of botulinum neurotoxin type A and its subunits has been carried out. The heavy and light chains of neurotoxin have the same ratios of polar and non-polar amino acids (1.3:1), the amount of tryptophan residues in the heavy chain is 4 times as much as that in the light chain, and the number of SH-groups exceeds that in ...

[Pharmacology of botulinum neurotoxin type A].

Wiener klinische Wochenschrift, 2004
The efficacies and potencies of Dysport and Botox, botulinum neurotoxin type A complexes approved for therapy, were investigated under various conditions. Conditions for maximal expression of biological activities of both commercial products were explored in vitro in the phrenic nerve-hemidiaphragm preparation whilst conditions for optimal distribution
