Interlaboratory performance testing on EPIC v2.0 CNS tumor profiling demonstrates high reproducibility of tumor classification but reveals the need for harmonized copy number variation reporting

Using the Infinium MethylationEPIC v2.0 array and Heidelberg Brain Tumor Classifier v12.8, 24 international laboratories achieved highly reproducible CNS tumor classification (97.9% correct; median β‐correlation r = 0.99), while copy number variation interpretation showed substantial interlaboratory variability, highlighting the need for harmonized CNV

Circumscribed Meningeal Melanocytic Neoplasms: CNS WHO Grade, Molecular Profile, and Clinical Outcomes

In this study including 31 circumscribed meningeal melanocytic neoplasms (melanocytoma, intermediate‐grade melanocytic tumor [IMT], and melanoma), overall survival (OS) differed significantly among the groups, with melanoma showing significantly worse OS than melanocytoma and IMT showing intermediate OS between melanocytoma and melanoma.

Gliomas with pleomorphic and pseudopapillary features (GPAP) are circumscribed tumors with targetable mutations, prolonged survival, and frequent tumor predisposition

DNA methylation profiling identifies GPAP as a distinct circumscribed glioma with a characteristic expansive, multicystic radiological appearance, pleomorphic and pseudopapillary histology, a recognizable CNV profile, druggable molecular alterations, and recurrent cancer predisposition syndromes.
