Functional Characterization of BRCA1 and BRCA2: Clues from Their Interacting Proteins

Journal of Mammary Gland Biology and Neoplasia, 1998
The familial breast and ovarian cancer susceptibility genes, BRCA1 and BRCA2 have been the subject of extensive functional analysis studies since their cloning. Clues to their biological role in maintaining the genomic integrity were provided by studies that revealed their interaction with the recombination repair protein HsRad51.

A Novel BRCA2-Binding Protein and Breast and Ovarian Tumorigenesis

New England Journal of Medicine, 2004
A fundamental question of cancer genetics is how genes responsible for inherited cancer contribute to the far more frequent cases of cancer that are purely somatic in origin.

Tetratricopeptide-motif-mediated interaction of FANCG with recombination proteins XRCC3 and BRCA2

DNA Repair, 2006
Fanconi anaemia is an inherited chromosomal instability disorder characterised by cellular sensitivity to DNA interstrand crosslinkers, bone-marrow failure and a high risk of cancer. Eleven FA genes have been identified, one of which, FANCD1, is the breast cancer susceptibility gene BRCA2. At least eight FA proteins form a nuclear core complex required

Impact of Disrupted Brca2 Protein-Protein Interactions on DNA Repair and Tumorigenesis

2001
Abstract : In this report, we have analyzed the protein encoded by the murine Brca2 locus. We find that murine Brca2 shares multiple properties with human BRCA2 including its regulation during the cell cycle, localization to nuclear foci, and interaction with Brca1 and Rad51.

Brca1 and Brca2 protein expression patterns in different tissues of murine origin

International Journal of Oncology, 2001
We have analyzed by immunohistochemistry Brca1 and Brca2 protein expression in mouse during embryonic development, in adult tissues, and during postnatal mammary gland development. Our observations confirm previous localization of Brca1 and Brca2 mRNA on frozen sections by in situ hybridization, and demonstrate that Brca1 and Brca2 proteins are ...

Biochemical Analysis of the BRCA2 Protein Complex

2004
Abstract : This linked career development award and idea award (CDA part) aims at relieving my administrative duties to acquire molecular biology skills in biology research. I have learned all the molecular biology techniques in accordance to Task 1 and 2 as stated in the Statement of Work.

The Brca1 and Brca2 proteins and tumor pathogenesis.

Anticancer research, 2000
Germline alterations of the BRCA1 or BRCA2 genes result in susceptibility to breast and ovarian cancer. Protein-protein interaction studies, transcription activity and mouse knockout experiments have suggested that the Brca1 and Brca2 proteins are of importance in DNA repair and maintenance of genome integrity, possibly due to the transactivation ...

Nuclear location and cell cycle regulation of the BRCA2 protein.

Cancer research, 1998
Women carrying a germ-line mutation in the BRCA1 or BRCA2 genes have a high risk of developing breast cancer, and loss of the wild-type allele in tumors suggests that these genes function as tumor suppressor genes. The BRCA2 gene encodes a 3418-amino acid protein with no significant sequence similarity to any known protein.
