The hyperactivation of PI3K/AKT signaling in PTEN wild‐type triple‐negative breast cancer represents a clinical paradox. We delineate a novel post‐translational regulatory axis wherein the oncogene TSPYL5 competitively antagonizes the deubiquitinase USP10.

This study identifies ARID3A as a key immunosuppressive transcription factor in TNBC. Its inhibition activates the type I IFN pathway, boosting CD8+ T cell infiltration and sensitizing tumors to anti‐PD‐1. The FDA‐approved migraine drug Rimegepant targets ARID3A, enhances immunotherapy efficacy in preclinical models, and establishes a druggable axis to

Prostate cancer is immunologically ‘cold’, with scarce, dysfunctional type 1 conventional dendritic cells (cDC1s) that limit T cell priming. We introduce an aptamer‐targeted liposomedelivering FMS‐like tyrosine kinase 3 ligand (Flt3L) and chlorin e6 (Ce6). Ultrasound induces antigen release and cDC1s recruitment, creating an in situ cDC1 vaccine.

Breast Cancer Surveillance Following Ovarian Cancer in BRCA Mutation Carriers
BRCA1 or 2 mutations result in higher cancer risk for breast cancer (BC) and epithelial ovarian cancer (EOC) for carriers than exists in the general population. Optimal breast imaging surveillance in these patients has not been well defined.

Risk-Reducing Bilateral Salpingo-Oophorectomy for BRCA Mutation Carriers and Hormonal Replacement Therapy: If It Should Rain, Better a Drizzle than a Storm [editorial]. [PDF]
Women carrying a BRCA mutation have an increased risk of developing breast and ovarian cancer. The most effective strategy to reduce this risk is the bilateral salpingo-oophorectomy, with or without additional risk-reducing mastectomy.

Germline breast cancer susceptibility gene mutations and breast cancer outcomes
Background It is unclear whether germline breast cancer susceptibility gene mutations affect breast cancer related outcomes. We wanted to evaluate mutation patterns in 20 breast cancer susceptibility genes and correlate the mutations with clinical ...

Topology‐Aware Deep Learning on Higher‐Order Structures for Drug Response Prediction
We present TopDr, a topology‐aware deep learning framework that encodes both drugs and cell lines as multiscale simplicial complexes, capturing interactions at the 0‐, 1‐, and 2‐simplex levels. By jointly integrating local higher‐order neighborhoods and global topological structures, TopDr generates enriched representations for sensitivity prediction ...
