In Vitro Biophysical Characterization of Candidalysin: A Fungal Peptide Toxin

2022
In 2016, the first peptide toxin in any human fungal pathogen was identified. It was discovered in Candida albicans and was named candidalysin. Candidalysin is an amphipathic cationic peptide that damages cell membranes. Like most lytic peptides, candidalysin shows alpha-helical secondary structure.

Heparin interacts with candidalysin and neutralizes its cytotoxicity to oral epithelial cells

Journal of Oral Biosciences, 2023
Candidalysin is a peptide toxin produced by Candida albicans that causes damage to epithelial cells by destabilizing the plasma membrane. This study aimed to evaluate heparin's ability to neutralize candidalysin and protect epithelial cells from lysis. The study was conducted using a human oral epithelial cell line and synthetic candidalysin.

Candidalysin: An unlikely aide for fungal gut commensalism

Cell Host & Microbe
Fungi colonize the mammalian gastrointestinal (GI) tract and can adopt both commensal and opportunistic lifestyles. In a recent issue of Nature, Liang et al. unraveled the complex interplay between Candida morphotypes and the gut bacterial microbiota and described a key role for candidalysin in gut colonization.1.

Secretion of the fungal toxin candidalysin is dependent on conserved precursor peptide sequences

Nature Microbiology
The opportunistic fungal pathogen Candida albicans damages host cells via its peptide toxin, candidalysin. Before secretion, candidalysin is embedded in a precursor protein, Ece1, which consists of a signal peptide, the precursor of candidalysin and seven non-candidalysin Ece1 peptides (NCEPs), and is found to be conserved in clinical isolates. Here we

Epithelial targets of Candidalysin

C57BL/6 or CD1 mice were administered vehicle (3% carboxymethylcelluose in PBS) gel or gel containing 5 mg/mL dextran sodium sulfate (DSS) 1 day prior to infection. They were then challenged intravaginally with 5x10^6 C. albicans followed by vehicle or DSS 2 h later, which continued daily throughout the study.

Candidalysin induces Tim-3 expression in macrophages to suppress antifungal immunity in oropharyngeal candidiasis

International Immunopharmacology
Oropharyngeal candidiasis significantly impacts patients' quality of life. Although the immune mechanisms underlying OPC have been explored, the roles of immune checkpoints and macrophages in this context remain unclear. We employed single-cell techniques to analyze changes in the oral immune microenvironment and explored the role of Tim-3 in OPC.

Detection of Cytolysis Peptide Toxin Gene (Candidalysin) in Candida albicans Isolates

Indian Journal of Public Health Research & Development, 2019
This study was launched to identify the presence of the gene; ece1 (extent of cell elongation 1), encodes for a precursor protein related to the candidalysin, a cytolytic peptide toxin important for the induction of mucosal-related infections, in isolates of Candida albicans. Initially, 30 isolates of C.
