Interactions of Casein Components with lmmobilized γ-Casein
γ-カゼインと他カゼイン成分の相互作用を固定化γ-カゼインを用いたアフィニティークロマトグラフィーにより調べた.全カゼインを1M NaClを含むpH7.0の0.05M燐酸緩衝液(I)に溶解し,γ-カゼインーセファロース4Bカラム(0.9×16cm)に添加した後,同緩衝液で溶出に,さらに,NaClを含まない同緩衝液,pH7.0(Ia), pH7.8(II),0.05Mトリス緩衝液,pH8.6(III),2Mおよび4M尿素を含む同緩衝液(IIIa, IIIb)で溶出した.得られた画分を4M尿素を含むディスク電気泳動により観察した.溶出温度30°Cでは,カゼイン成分は緩衝液Iでカラムに吸着されたが,20°Cでは一部吸着されずに溶出し,2°Cでは吸着された量が少なく,温度により著しく影響された.20 ...
