Caspase-2S relative mRNA expression levels in neurological disease.
(A) Caspase-2S mRNA expression levels trend lower in AD but not MCI compared to CN controls. Caspase-2S mRNA do not differ in LBD (B) or HD (C) compared to controls.

Aging Is a Key Driver for Adult Acute Myeloid Leukemia
Acute myeloid leukemia (AML) is a classical age‐related hematologic malignancy, and a key driver of AML is aging, which profoundly regulates intrinsic factors such as genomic instability, epigenetic reprogramming, and metabolic dysregulation, and alters bone marrow microenvironment.

Glutathione peroxidase 8 negatively regulates caspase‐4/11 to protect against colitis
Human caspase‐4 and its mouse homolog caspase‐11 are receptors for cytoplasmic lipopolysaccharide. Activation of the caspase‐4/11‐dependent NLRP3 inflammasome is required for innate defense and endotoxic shock, but how caspase‐4/11 is modulated remains ...

Mutant NPM1 in Acute Myeloid Leukemia Initiation and Maintenance
NPM1 mutations drive acute myeloid leukemia by acting as neomorphic transcriptional regulators that cooperate with Menin–MLL and XPO1 to sustain HOX/MEIS1 expression and block differentiation. Targeting these mutant‐specific transcriptional dependencies provides a rational therapeutic strategy for NPM1‐mutated AML.

An Induced Proximity Model for Caspase-8 Activation [PDF]
The assembly of the CD-95 (Fas/Apo-1) receptor death-inducing signaling complex occurs in a hierarchical manner; the death domain of CD-95 binds to the corresponding domain in the adapter molecule Fas-associated death domain (FADD) Mort-1, which in turn recruits the zymogen form of the death protease caspase-8 (FLICE/Mach-1) by a homophilic interaction

ABSTRACT Objective Super‐Refractory Status Epilepticus (SRSE) is a rare, life‐threatening neurological emergency with unclear etiology in many cases. Mitochondrial dysfunction, often due to disease‐causing genetic variants, is increasingly recognized as a cause, with each gene producing distinct pathophysiological mechanisms.

Lymphocyte life and death: The caspase-8 connection [PDF]
Caspase-8 plays an important role in death receptor-mediated apoptosis. In contrast to its apoptotic function, recent studies have suggested caspase-8 also plays a role in the immune response, as caspase-8 is essential for lymphocyte proliferation in response to antigen stimulation.

Cathepsin D primes caspase-8 activation by multiple intra-chain proteolysis
During the resolution of inflammatory responses, neutrophils rapidly undergo apoptosis. A direct and fast activation of caspase-8 by cathepsin D was shown to be crucial in the initial steps of neutrophil apoptosis.
