Caspase-9 as a therapeutic target for treating cancer

Expert Opinion on Therapeutic Targets, 2015
Caspase-9 is the apoptotic initiator protease of the intrinsic or mitochondrial apoptotic pathway, which is activated at multi-protein activation platforms. Its activation is believed to involve homo-dimerization of the monomeric zymogens. It binds to the apoptosome to retain substantial catalytic activity.

The role of caspase‐8, caspase‐9, and apoptosis inducing factor in periodontal disease

The Journal of Periodontology, 2018
BACKGROUND Caspases are key mediators of apoptosis. Caspase-8 mediates extrinsic, and caspase-9 initiates the intrinsic pathway of apoptosis. Apoptosis Inducing Factor (AIF), a mitochondrial proapoptotic protein, mediates cell death by a caspase ...

Expression of phosphorylated caspase‐9 in gastric carcinomas

APMIS, 2007
Alterations of caspases, the main executioners of apoptosis, have been described in human cancers. Caspase‐9 plays a crucial role in the initiation phase of the intrinsic apoptosis pathway. Caspase‐9 is phosphorylated at Thr125 through the mitogen‐activated protein kinase (MAPK) pathway, and this phosphorylation is associated with inhibition of caspase‐

cDNA cloning and promoter analysis of rat caspase-9

Biochemical Journal, 2001
Caspase-9 is the apex caspase of the mitochondrial pathway of apoptosis, which plays a critical role in apoptotic initiation and progression. However, gene regulation of caspase-9 is largely unknown. This is in part due to the lack of information on the gene promoter.

Caspase-9 activation in hypoxic human corneal epithelial cells

Apoptosis, 2003
The purpose of this study was to determine the effect of hypoxia on caspase-8 and -9 gene and protein expression and activity in corneal epithelium. Non-transformed human corneal epithelial cells (HCEC) were cultured in 2% oxygen. A cDNA expression array coupled with densitometric analysis was used to compare relative mRNA expression levels of 96 ...

Cytochrome C and Caspase-9 Expression in Huntington 's Disease

NeuroMolecular Medicine, 2002
There is increasing evidence implicating apoptosis-mediated cell death in the pathogenesis of neurodegenerative diseases. One important event in the apoptotic cascade is the release of cytochrome c by mitochondria into the cytoplasm, activating caspase-9, leading to the subsequent activation of downstream executioner caspases.
