A new crystal form of Aspergillus oryzae catechol oxidase and evaluation of copper site structures in coupled binuclear copper enzymes. [PDF]

open access: yesPLoS ONE, 2018
Coupled binuclear copper (CBC) enzymes have a conserved type 3 copper site that binds molecular oxygen to oxidize various mono- and diphenolic compounds.

Some properties of active and latent catechol oxidase of mushroom

open access: yesActa Agrobotanica, 2013
Latent form of mushroom catechol oxidase was activated by O,1% sodium dodecyl sulfate (SDS). Catalytic power of the latent form, calculated from the kinetic parameters was 1,8 times higher than that of active one.

Aurone synthase is a catechol oxidase with hydroxylase activity and provides insights into the mechanism of plant polyphenol oxidases [PDF]

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2016
Tyrosinases and catechol oxidases belong to the family of polyphenol oxidases (PPOs). Tyrosinases catalyze the o-hydroxylation and oxidation of phenolic compounds, whereas catechol oxidases were so far defined to lack the hydroxylation activity and ...

A Novel Catechol Oxidase Enzyme Electrode for the Specific Determination of Catechol

open access: yesBioscience, Biotechnology, and Biochemistry, 1998
An enzyme electrode for the specific determination of catechol was developed by using catechol oxidase (EC 1.10.3.1) from eggplant (Solanum melangena L.) in combination with a dissolved oxygen probe. Optimization studies of the prepared catechol oxidase enzyme electrode established a phosphate buffer 50 mM at pH 7.0 and 35°C to provide the optimum ...

The Binding Affinity of Small Molecules with Yam Tyrosinase (Catechol Oxidase): A Biophysical Study [PDF]

open access: yesBiochemistry Research International, 2019
Yam tyrosinase has become an economically essential enzyme due to its ease of purification and abundant availability of yam tubers. However, an efficient biochemical and biophysical characterization of yam tyrosinase has not been reported. In the present

Conversion of walnut tyrosinase into a catechol oxidase by site directed mutagenesis. [PDF]

open access: yesSci Rep, 2020
AbstractPolyphenol oxidases (PPOs) comprise tyrosinases (TYRs) and catechol oxidases (COs), which catalyse the initial reactions in the biosynthesis of melanin. TYRs hydroxylate monophenolic (monophenolase activity) and oxidize diphenolic (diphenolase activity) substrates, whereas COs react only with diphenols.

Structural and spectroscopic studies of a model for catechol oxidase [PDF]

open access: yesJBIC Journal of Biological Inorganic Chemistry, 2008
A binuclear copper complex, [Cu2(BPMP) (OAc)2][ClO4] x H2O, has been prepared using the binucleating ligand 2,6-bis[bis(pyridin-2-ylmethylamino)methyl]-4-methylphenol (H-BPMP). The X-ray crystal structure reveals the copper centers to have a five-coordinate square pyramidal geometry, with the acetate ligands bound terminally.

Catechol oxidase activity of comparable dimanganese and dicopper complexes

open access: yesDalton Transactions, 2018
Dicopper complexes proved to be more reactive catechol oxidase mimics when compared to dimanganese complexes supported by identical ligand frameworks.

Purification and Enzymatic Properties of Soluble and Membrane-Bound Polyphenol Oxidase from Agaricus bisporus [PDF]

open access: yesShipin Kexue, 2023
An extraction and purification method for soluble polyphenol oxidase (sPPO) and membrane-bound polyphenol oxidases (mPPO) from Agaricus bisporus was developed involving temperature-induced phase partitioning, fractional precipitation with ammonium ...
