A series of novel piperidamide-3-carboxamide derivatives were synthesized and evaluated for their inhibitory activities against cathepsin K. Among these derivatives, compound H-9 exhibited the most potent inhibition, with an IC50 value of 0.08 µM ...

Non-invasive in vivo imaging of tumour-associated cathepsin B by a highly selective inhibitory DARPin [PDF]
Cysteine cathepsins often contribute to cancer progression due to their overexpression in the tumour microenvironment and therefore present attractive targets for non-invasive diagnostic imaging. However, the development of highly selective and versatile

Zmat1 deficiency mitigates pathological bone loss by impairing osteoclastogenesis and promoting osteoblastogenesis. Mechanistically, in osteoclasts, Zmat1 loss relieves transcriptional repression of the E3 ligase TRIM46, promoting YAP1 degradation and inhibiting osteoclastogenic genes.

Numerous studies have linked cathepsins and their inhibitor cystatin C to tumor invasion and metastasis. We examined whether cathepsin B, cathepsin H, cathepsin X and cystatin C could be detected in sera from women with early-stage or inflammatory breast

Producing MSCs on rigid culture substrates induces a scar‐making phenotype, jeapordizing therapeutic success. ‘Tissue‐soft’ surfaces prevent MSC fibrogenesis and preserve regenerative traits. An epigenetic network, driven by HOXA11 and SALL1, maintains ‘soft memory’ by keeping chromatin open in relaxed MSCs, promoting anti‐fibrotic programs.

Peptide dendrimer–based nanoplatform with mitochondria‐targeting capability was developed for synergistic chemo‐photothermal therapy. The system promotes efficient transcytosis, induces mitochondrial dysfunction, triggers immunogenic cell death (ICD), and enhance antitumor immunity by promoting dendritic cell maturation and T‐cell activation, resulting

A bioavailable cathepsin S nitrile inhibitor abrogates tumor development [PDF]
BACKGROUND: Cathepsin S has been implicated in a variety of malignancies with genetic ablation studies demonstrating a key role in tumor invasion and neo-angiogenesis.

ABSTRACT The muscle capsule of Trichinella is a critical structure that impedes immune attacks and drug penetration, yet the molecular mechanisms underlying its formation remain poorly understood. Using a high‐quality super‐pangenome comprising 12 Trichinella species, we compared extensive genomic variations between encapsulating and non‐encapsulating ...
