Cdt1 and Cdc6 Are Destabilized by Rereplication-induced DNA Damage [PDF]
The replication factors Cdt1 and Cdc6 are essential for origin licensing, a prerequisite for DNA replication initiation. Mechanisms to ensure that metazoan origins initiate once per cell cycle include degradation of Cdt1 during S phase and inhibition of Cdt1 by the geminin protein.

Mismatch repair regulates Cdt1 after UV damage [PDF]
Mismatch repair (MMR) corrects mismatches in newly synthesized DNA strands.1 Key factors in the recognition of the mismatched bases are the Msh2 and Msh6 ATPases, which act during DNA replication a...

Backgrounds. Diagnostic markers of extraintestinal infection in Escherichia coli (E. coli) remain unclear in the literature. Extraintestinal pathogenic E. coli (ExPEC) is differentiated from other E.

Distinct and sequential re-replication barriers ensure precise genome duplication.
Achieving complete and precise genome duplication requires that each genomic segment be replicated only once per cell division cycle. Protecting large eukaryotic genomes from re-replication requires an overlapping set of molecular mechanisms that prevent

DDB1 Maintains Genome Integrity through Regulation of Cdt1 [PDF]
DDB1, a component of a Cul4A ubiquitin ligase complex, promotes nucleotide excision repair (NER) and regulates DNA replication. We have investigated the role of human DDB1 in maintaining genome stability. DDB1-depleted cells accumulate DNA double-strand breaks in widely dispersed regions throughout the genome and have activated ATM and ATR cell cycle ...

MCM9 Binds Cdt1 and Is Required for the Assembly of Prereplication Complexes [PDF]
Prereplication complexes (pre-RCs) define potential origins of DNA replication and allow the recruitment of the replicative DNA helicase MCM2-7. Here, we characterize MCM9, a member of the MCM2-8 family. We demonstrate that MCM9 binds to chromatin in an ORC-dependent manner and is required for the recruitment of the MCM2-7 helicase onto chromatin.

In Vivo DNA Re-replication Elicits Lethal Tissue Dysplasias
Summary: Mammalian DNA replication origins are “licensed” by the loading of DNA helicases, a reaction that is mediated by CDC6 and CDT1 proteins. After initiation of DNA synthesis, CDC6 and CDT1 are inhibited to prevent origin reactivation and DNA ...

Geminin Stabilizes Cdt1 during Meiosis in Xenopus Oocytes [PDF]
During the mitotic cell cycle, Geminin can act both as a promoter and inhibitor of initiation of DNA replication. As a promoter, Geminin stabilizes Cdt1 and facilitates its accumulation leading to the assembly of the pre-replication complex on DNA. As an inhibitor, Geminin prevents Cdt1 from loading the mini-chromosome maintenance complex onto pre ...
