Cellulose, cellulases and cellulosomes
Current Opinion in Structural Biology, 1998The structural complexity and rigidity of cellulosic substrates have given rise to a phenomenal diversity of degradative enzymes--the cellulases. Cellulolytic microorganisms produce a wide variety of different catalytic and noncatalytic enzyme modules, which form the cellulases and act synergistically on their substrate. In some microbes, several types

Deglycosylation of cellulosomal enzyme enhances cellulosome assembly in Saccharomyces cerevisiae
Journal of Biotechnology, 2012We have estimated the effects of hyper-mannosylation of dockerin-type cellulase on cellulosome assembly by using Saccharomyces cerevisiae and 44 protein glycosylation mutants, because the heterologous protein displayed on yeast is assumed to be modified by yeast-specific hyper-mannosylation.

The cellulosome — A treasure-trove for biotechnology
Trends in Biotechnology, 1994The cellulases of many cellulolytic bacteria are organized into discrete multienzyme complexes, called cellulosomes. The multiple subunits of cellulosomes are composed of numerous functional domains, which interact with each other and with the cellulosic substrate.

Carbohydrate Depolymerization by Intricate Cellulosomal Systems
2023Cellulosomes are multi-enzymatic nanomachines that have been fine-tuned through evolution to efficiently deconstruct plant biomass. Integration of cellulosomal components occurs via highly ordered protein-protein interactions between the various enzyme-borne dockerin modules and the multiple copies of the cohesin modules located on the scaffoldin ...

Polysaccharides derived from lignocellulose are promising sustainable carbon sources. Cellulosome is a supramolecular machine integrating multi-function enzymes for effective lignocellulose bio-saccharification. However, how various non-cellulose components of lignocellulose affect the cellulosomal saccharification is hitherto unclear. This study first

Cellulosomes: Highly Efficient Cellulolytic Complexes
2020Cellulosomes are elaborate multienzyme complexes capable of efficiently deconstructing lignocellulosic substrates, produced by cellulolytic anaerobic microorganisms, colonizing a large variety of ecological niches. These macromolecular structures have a modular architecture and are composed of two main elements: the cohesin-bearing scaffoldins, which ...

THE CELLULOSOME: The Exocellular Organelle of Clostridium
Annual Review of Microbiology, 1993The cellulolytic enzyme complex of the anaerobic thermophile Clostridium thermocellum is reviewed. This complex, called the cellulosome, is cell associated and has a mass of from 2 x 10(6) to 6.5 x 10(6) Daltons. It consists of from 14 to 26 different polypeptides.

Cellulosome and noncellulosomal cellulases of Clostridium cellulovorans
Extremophiles, 1998This paper reviews the properties of the cellulosome and noncellulosome cellulases produced by Clostridium cellulovorans, an anaerobic, mesophilic, spore-forming microorganism that produces copious amounts of cellulase. The three major subunits of the cellulosome, CbpA, exoglucanase S (ExgS), and P100, are described, as well as the properties of the ...

The Cellulosome of Clostridium thermocellum
1988Publisher Summary The accelerated interest in microbial cellulases stems from their potential industrial application. This area is an excellent example in which basic and applied science are closely interlinked: the better understood the mechanism of cellulase action; the better can be the applicative value as a biotechnological process. This chapter
