The Microtubule Binding Properties of CENP-E's C-Terminus and CENP-F [PDF]
CENP-E (centromere protein E) and CENP-F (centromere protein F), also known as mitosin, are large, multi-functional proteins associated with the outer kinetochore. CENP-E features a well-characterized kinesin motor domain at its N-terminus and a second microtubule-binding domain at its C-terminus of unknown function.

Swapping CENP-A at the centromere
Nature Cell Biology, 2013Faithful genome segregation depends on the functions of the eukaryotic centromere, which is characterized by the histone variant CENP-A. Gene replacement in human cells and fission yeast has now been used to show how CENP-A biochemically encodes centromere identity, as well as reveal an unexpected role for CENP-B in centromere function.

Live‐cell imaging reveals sustained centromere binding of CENP‐T via CENP‐A and CENP‐B
Journal of Biophotonics, 2008AbstractAt the centromere, a network of proteins, the kinetochore, assembles in order to grant correct chromatin segregation. In this study the dynamics and molecular interactions of the inner kinetochore protein CENP‐T were analyzed employing a variety of fluorescence microscopy techniques in living human cells.

Putting CENP-A in its place [PDF]
The centromere is the chromosomal region that directs kinetochore assembly during mitosis in order to facilitate the faithful segregation of sister chromatids. The location of the human centromere is epigenetically specified. The presence of nucleosomes that contain the histone H3 variant, CENP-A, are thought to be the epigenetic mark that indicates ...

HJURP inhibits proliferation of ovarian cancer cells by regulating CENP-A/CENP-N
Bulletin du Cancer, 2022High expression of Holliday Junction-Recognizing Protein (HJURP) has been shown to be a marker of poor prognosis in ovarian cancer. The objective of this study was to investigate the molecular mechanisms of HJURP in ovarian cancer (OC) progression.Gene Expression Profiling Interactive Analysis (GEPIA) was used to analyze the gene expression profile ...

CENP-K and CENP-H may form coiled-coils in the kinetochores
Science in China Series C: Life Sciences, 2009Kinetochores are large proteinaceous structure on the surface of chromosomes' primary constriction during mitosis. They link chromosomes to spindle microtubules and also regulate the spindle assembly checkpoint, which is crucial for correct chromosome segregation in all eukaryotes. The better known core networks of kinetochores include the KMN network (

Mass Spectrometry Analysis to Identify Ubiquitylation of EYFP-tagged CENP-A (EYFP-CENP-A)
Journal of Visualized Experiments, 2020Studying the structure and the dynamics of kinetochores and centromeres is important in understanding chromosomal instability (CIN) and cancer progression. How the chromosomal location and function of a centromere (i.e., centromere identity) are determined and participate in accurate chromosome segregation is a fundamental question.

Epigenetic specification of centromeres by CENP-A
Experimental Cell Research, 2009Centromeres are the chromosomal loci that direct the formation of the kinetochores. These macromolecular assemblies mediate the interaction between chromosomes and spindle microtubules and thereby power chromosome movement during cell division. They are also the sites of extensive regulation of the chromosome segregation process.

Development of a CENP‐A/CENP‐B–specific immune response in a patient with systemic sclerosis
Arthritis & Rheumatism, 2002AbstractAntibodies directed against an epitope motif on CENP‐A have been shown to cross‐react with mimotopes on other autoantigens and on Epstein‐Barr nuclear antigen 1 (EBNA‐1), suggesting a molecular mimicry. We describe here the gradual development of an anticentromere immune response in a patient with systemic sclerosis, which started from an ...

Assembly of the Inner Kinetochore Proteins CENP‐A and CENP‐B in Living Human Cells
ChemBioChem, 2007AbstractDNA segregation in mammalian cells during mitosis is an essential cellular process that is mediated by a specific subchromosomal protein complex, the kinetochore. Malfunction of this complex results in aneuploidy and can cause cancer. A subkinetochore complex, the “inner kinetochore”, is present at the centromere during the entire cell cycle ...
