Enzymatic Formation of Unnatural Aromatic Polyketides by Chalcone Synthase

Biochemical and Biophysical Research Communications, 2000
Substrate specificity of recombinant chalcone synthase (CHS) from Scutellaria baicalensis (Labiatae) was investigated using chemically synthesized aromatic and aliphatic CoA esters. It was demonstrated for the first time that CHS converted benzoyl-CoA to phlorobenzophenone (2,4,6-trihydroxybenzophenone) along with pyrone by-products. On the other hand,

Organization of the genes encoding chalcone synthase in Pisum sativum

Plant Molecular Biology, 1993
To analyze the regulation of defense-related genes by signal molecules produced by phytopathogens, we isolated genes that encode chalcone synthase (CHS) in Pisum sativum. We have obtained seven independent genomic clones that contain at least seven classes of CHS genes, identified by the hybridization analysis to CHS cDNA and by the restriction mapping

In Trifolium subterraneum, chalcone synthase is encoded by a ultigene family

Gene, 1994
Chalcone synthase (CHS) catalyzes the first and key regulatory step in flavonoid biosynthesis. We report the existence and characterization of a CHS multigene family present in Trifolium subterraneum L. cultivar Karridale. The CHS family consists of at least four members, which are tightly clustered in a 15-kb region.

Engineered Biosynthesis of Plant Polyketides:  Manipulation of Chalcone Synthase

Organic Letters, 2006
[reaction: see text]. Chalcone synthase (CHS) is a plant-specific type III polyketide synthase catalyzing condensation of 4-coumaroyl-CoA with three molecules of malonyl-CoA. Surprisingly, it was demonstrated that S338V mutant of Scutellaria baicalensis CHS produced octaketides SEK4/SEK4b from eight molecules of malonyl-CoA.

Structure, Function, and Regulation of the Chalcone Synthase

1993
Publisher Summary This chapter presents an overview of the activity, function, and control of chalcone synthase (CHS) enzyme. It describes the reaction it catalyzes, the secondary metabolic products of the pathways in which it is involved, and the functions of these products.

Divergent evolution of the thiolase superfamily and chalcone synthase family

Molecular Phylogenetics and Evolution, 2008
Enzymes of the thiolase superfamily catalyze the formation of carbon-carbon bond via the Claisen condensation reaction. Thiolases catalyze the reversible non-decarboxylative condensation of acetoacetyl-CoA from two molecules of acetyl-CoA, and possess a conserved Cys-His catalytic diad.

Structure and function of the chalcone synthase superfamily of plant type III polyketide synthases

Natural Product Reports, 2010
Covering: 2002 to 2009 This review covers recent advances in structure and function studies on the chalcone synthase (CHS) superfamily of plant type III polyketide synthases (PKSs), which catalyze iterative decarboxylative condensations of malonyl unit with a CoA-linked starter molecule to produce structurally diverse, pharmaceutically ...

Structure of the chalcone synthase gene of Antirrhinum majus

Molecular and General Genetics MGG, 1986
Genomic and cDNA clones of the chalcone synthase gene (chs) of Antirrhinum majus have been isolated and sequenced. By sequence comparison the structure of the gene was deduced. The coding region of the chs gene is interupted by two intervening sequences of 721 bp and 211 bp. The gene codes for a protein with a MW of 42,665 daltons.

[Cloning and characterization of chalcone synthase and chalcone isomerase genes in Arisaema heterophyllum].

Zhongguo Zhong yao za zhi = Zhongguo zhongyao zazhi = China journal of Chinese materia medica, 2019
Chalcone synthase( CHS) and chalcone isomerase( CHI) are key enzymes in the biosynthesis pathway of flavonoids. In this study, unigenes for CHS and CHI were screened from the transcriptome database of Arisaema heterophyllum. The open reading frame( ORFs) of chalcone synthase( Ah CHS) and chalcone isomerase( Ah CHI) were cloned from the plant by RT-PCR ...

Variability of Chalcone Synthase in Chamomile Accessions (Matricaria chamomilla)

Planta Medica
AbstractChamomile (Matricaria chamomilla) is an important medicinal plant whose beneficial activities partly rely on certain flavonoids. The first dedicated step in flavonoid biosynthesis is chalcone synthase (CHS, EC 2.3.1.74). The genomic DNA of CHS was studied in six chamomile specimens from different genotypes to describe interspecimen, as well as ...
