Galactokinase 1 Positively Regulates Mitochondrial Respiration by Phosphorylating TIMM13 as a Protein Kinase

open access: yesAdvanced Science, EarlyView.
GALK1 acts as a protein kinase beyond the phosphorylation of galactose. GALK1 phosphorylates TIMM13 at Y73 in the cytoplasm. This phosphorylation prevents premature oxidative folding of TIMM13 and ensures its entrance into the intermembrane space of mitochondrion, positively regulating mitochondrial respiration.

Pharmacological Inhibition of Hsp70 to Degrade c-FLIP: A Novel Indirect Approach for Cancer Therapy: A Narrative Review

open access: yesKurdistan Journal of Applied Research
The cellular FLICE-inhibitory protein (c-FLIP) is a master regulator of pro-grammed cell death, functioning as a key suppressor of extrinsic apoptosis mediated by death receptor signaling. Its persistent overexpression is a hall-mark of numerous cancers,

USP4‐Dependent CHAF1B Stabilization Regulates Distinct SETDB1 Ubiquitin States Linked to AKT T308 Signaling and Lipogenic Remodeling in HCC

open access: yesAdvanced Science, EarlyView.
USP4 stabilizes CHAF1B by limiting K48‐linked ubiquitination. Stabilized CHAF1B supports UHRF1‐associated K63‐linked SETDB1 ubiquitination and cytoplasmic redistribution, whereas CHAF1B loss favors VHL‐dependent K11‐associated degradative ubiquitination and proteasomal loss of SETDB1.

Lipid Metabolic Dysregulation Driven by OSMR Mutations Underlies Amyloidogenesis in Primary Localized Cutaneous Amyloidosis

open access: yesAdvanced Science, EarlyView.
OSMR mutations disrupt STAT5 signaling and AKR1B10‐mediated lipid metabolism, leading to lipid accumulation and protein aggregation in keratinocytes. This metabolic reprogramming drives amyloid deposition in primary localized cutaneous amyloidosis, revealing a mechanistic link between lipid dysregulation and amyloidogenesis.

Microglia-derived nanovesicles synchronize macroautophagy and chaperone-mediated autophagy for Alzheimer’s disease therapy

open access: yesSignal Transduction and Targeted Therapy
Dysregulated autophagy is a hallmark of Alzheimer’s disease (AD), yet the extent of impairment in macroautophagy and chaperone-mediated autophagy (CMA) remains unclear.

Therapeutic landscape of Fabry disease: advances and challenges from classical strategies to emerging therapies

open access: yesFrontiers in Medicine
Fabry disease (FD), as an X-linked lysosomal storage disorder (LSD), has seen significantly improved in patient prognosis since enzyme replacement therapy (ERT) was applied clinically in 2001. However, ERT has drawbacks such as immunogenicity, individual
