Targeted chimeric degraders engage extracellular or membrane proteins and endocytosable cell‐surface receptors, forming ternary complexes that induce receptor‐mediated internalization. Following endocytosis, post‐endocytic sorting—not internalization alone—determines target fate.

Collagen‐Binding IL‐15 Immunocytokine Armed Oncolytic Adenovirus Establishes a Self‐Reinforcing Immunostimulatory Milieu to Potentiate Antitumor Immunity

Repurposing the dense tumor stroma from a physical barrier into an anchoring scaffold, an engineered oncolytic adenovirus encoding IL15C‐CBD overcomes the immunosuppressive microenvironment in triple‐negative breast cancer. Intratumoral viral replication secretes this collagen‐anchored immunocytokine, forming a localized cytokine depot.

Anti-CD19 chimeric antigen receptor targeting of CD19 + acute myeloid leukemia

Aberrant expression of CD19 in acute myeloid leukemia (AML) is commonly associated with t(8;21)(q22;q22), although AML cases lacking this translocation occasionally express CD19. Mixed-phenotype acute leukemia also frequently expresses CD19.

A Metabolic‐Epigenetic Axis of AARS1/IGF2BP3‐K280 Lactylation Sustains Endometrial Cancer Stemness and Recurrence

Lactate‐driven endometrial cancer relapse is mediated by enzymatic protein modification. Elevated lactate levels activate AARS1, which catalyzes K280 lactylation of IGF2BP3, therefore stabilizing HMGA2 mRNA and promoting tumor stemness and recurrence.
