Rapid detection of chitosanase activity in chitosanase gene-transformed strains of Enterobacter cloacae by lytic infection of specific bacteriophages

In combination with lytic infection by virulent phages, a simple method for monitoring transgenic strains of Enterobacter cloacae was developed in this study. First, 15 strains of E. cloacae were used as indicator bacteria to isolate virulent phages with different host ranges.

This communication reports the deposit of Streptomyces No. 6 in the National Collection of Industrial Bacteria, Aberdeen, AB2 1RY, Scotland, UK (Accession number 13037) and gives some references on its use in lysing cell walls of fungi in the Mucorales leading to protoplast formation. Its use in biotechnological fields is also referred to.

Chitosanase from the culture filtrate of Nocardia orientalis was purified to apparent homogeneity by precipitation with ammonium sulfate followed by CM-Sephadex chromatography, biospecific affinity chromatography on a Sepharose CL-4B with immobilized chitotriose and by gel filtration on Sephadex G-75.

Characterization of a Chitosanase Isolated from a Commercial Ficin Preparation

A chitosanolytic enzyme was purified from a commercial ficin preparation by affinity chromatographic removal of cysteine protease on pHMB-Sepharose 4B and cystatin-Sepharose 4B and gel filtration on Superdex 75 HR. The purified enzyme exhibited both chitinase and chitosanase activities, as determined by SDS-PAGE and gel activity staining.
