CDG due to Defective Membrane Transporters: Update

ABSTRACT Congenital disorders of glycosylation are genetic defects in the glycoprotein and glycolipid glycan assembly and attachment. Some 200 CDG have been reported since the first clinical description in 1980. Most CDG are enzymatic deficiencies, but 13 (6.5%) are defects in the ER, Golgi apparatus (GA), and plasma membrane transporters.

This study investigated if the epididymal amyloid CRES (cystatin‐related epididymal spermatogenic) is present in the male and female mammalian brain. CRES is expressed by hippocampal astrocytes, CA2 and CA3 neurons, and exhibits amyloid properties in the extracellular matrix.

Nanoparticle‐driven nose‐to‐brain drug delivery offers a noninvasive approach to bypass the blood–brain barrier (BBB) and directly modulate reactive astrocytes in CNS disorders. This review highlights biodegradable nanoparticles for astrocyte modulation, enhancing drug bioavailability, sustained release, and neuroinflammation control.
