High-intensity UV laser ChIP-seq for the study of protein-DNA interactions in living cells
Chromatin immunoprecipitation followed by DNA sequencing (ChIP-seq) can map transcription factor binding to a given genome. Here, Steube and colleagues devise a ChIP-seq technique based on photochemical crosslinking by high-intensity ultraviolet laser ...

This study demonstrates that iron overload triggers widespread chromatin compaction and transcriptional repression in human granulosa cells, recapitulating features of endometriosis. The epigenetic reprogramming is orchestrated by a TFEB‐SOX4‐SWI/SNF axis, with SOX4 acting as a central, dosage‐sensitive regulator.

Cigarette smoke accelerates the aging of immune cells in the lung. By combining human single‐cell sequencing, cell culture, and mouse models, the authors show that the protein GEM drives mitochondrial damage and senescence in alveolar macrophages by suppressing SIRT3.

Microfluidic systems and methods for chromatin immunoprecipitation (ChIP)
An integrated microfluidic chromatin immunoprecipitation assay dramatically improves the collection efficiency of ChIP DNA from cells. Immunoprecipitation of chromatin fragments is conducted in a microfluidic chamber with a large fraction of its volume ...

ANKS1B in the nucleus accumbens plays a critical role in the transition from controlled to escalated cocaine intake. Mechanistically, ANKS1B interacts with CBP to epigenetically suppress FoxO3 through H3K27 acetylation. The ANKS1B‐CBP‐FoxO3 signaling cascade presents a novel theraputic target for the treatment of cocaine addiction.

CAST-ChIP Maps Cell-Type-Specific Chromatin States in the Drosophila Central Nervous System
Chromatin organization and gene activity are responsive to developmental and environmental cues. Although many genes are transcribed throughout development and across cell types, much of gene regulation is highly cell-type specific.
