Crosstalk Between CTSB+ Glioblastoma Cells and S100A10+ Macrophages: A Self‐Reinforcing Circuit Promotes Immune Evasion and Limits Response to Immunotherapy

open access: yesAdvanced Science, EarlyView.
In glioblastoma, M2‐polarized macrophages secrete IL‐6, which activates STAT3 signaling in tumor cells to upregulate CTSB. Tumor‐derived CTSB binds the C‐terminus of macrophage S100A10, reinforcing M2 polarization and further IL‐6 secretion, thereby establishing a feedforward IL‐6/STAT3/CTSB/S100A10 loop. This cascade drives tumor growth, invasion, and

qChIP-MS reveals the local chromatin composition by label-free quantitative proteomics

open access: yesNature Communications
Chromatin immunoprecipitation (ChIP) has been a cornerstone for epigenetic analyses over the last decades, but even coupled to sequencing approaches (ChIP-seq), it is ultimately limited to one protein at a time. In a complementary effort, we here combine

Rhoifolin Alleviates Ulcerative Colitis by Targeting NMNAT1 and Activating SIRT1‐FOXO1 Signaling to Enhance ILC3s Effector Function

open access: yesAdvanced Science, EarlyView.
Rhoifolin directly targets NMNAT1, thereby activating nicotinamide salvage pathway and promoting intracellular NAD+ biosynthesis. Elevated NAD+ levels enhance SIRT1 activity, leading to FOXO1 deacetylation and nuclear translocation. Nuclear FOXO1 subsequently binds to the IL‐22 promoter, upregulating IL‐22 transcription and potentiating ILC3 effector ...

PAT-ChIP coupled with laser microdissection allows the study of chromatin in selected cell populations from paraffin-embedded patient samples [PDF]

open access: yes, 2014
Background: The recent introduction of pathology tissue-chromatin immunoprecipitation (PAT-ChIP), a technique allowing chromatin immunoprecipitation from formalin-fixed and paraffin-embedded (FFPE) tissues, has expanded the application potential of ...

Protocol for in vivo chromatin immunoprecipitation on purified chromatin isolated from mouse liver nuclei

open access: yesSTAR Protocols
Summary: Chromatin immunoprecipitation (ChIP) is used to investigate genome binding by transcription factors, but it can be problematic. We present a protocol to isolate fixed DNA-protein complexes from mouse liver prior to chromatin shearing.

Mepce Restrains Ferroptosis in Prostate Cancer Through 7SK–P‐TEFb Pause Control

open access: yesAdvanced Science, EarlyView.
Genome‐wide CRISPR screening identifies MEPCE as a pause‐control regulator that restrains ferroptosis in prostate cancer. MEPCE loss releases P‐TEFb/CDK9 from 7SK snRNP control, enhances RNAPII Ser2 phosphorylation, and accelerates NRF2‐HMOX1‐driven labile iron accumulation and lipid peroxidation, revealing a transcriptional kinetic gate that lowers ...

Quantitative ChIP-seq by Adding Spike-in from Another Species

open access: yesBio-Protocol, 2018
Chromatin immunoprecipitation followed by sequencing (ChIP-seq) is a routine procedure in the lab; however, epigenome-wide quantitative comparison among independent ChIP-seq experiments remains a challenge.

GM‐CSF Promotes Neutrophil PD‐L1 Expression Through the VEGFR‐2/STAT6/CSF2 Signaling Axis in Oral Squamous Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
VEGFR‐2 signaling in OSCC activates Src–STAT6‐dependent CSF2 transcription, driving tumor‐derived GM‐CSF secretion. GM‐CSF programs neutrophils to express PD‐L1 through STAT5–mTOR/S6K signaling, suppressing cytotoxic CD8+ T cells. This pathway reveals a tumor–neutrophil immune checkpoint circuit that limits anti‐PD‐1 responsiveness in OSCC.
