Alkaline serine protease was purified to homogeneity from culture supernatant of a thermophilic, alkaliphilic Bacillus sp. by 80% ammonium sulphate precipitation followed by CM-cellulose and DEAE-cellulose ion exchange column chromatography.

DEAE-Cellulose-Urea Chromatography of Casein in the Presence of 2-Mercaptoethanol

Abstract κ -Casein, after reduction of the disulfide bond(s) by 2-inercaptoethanol, can be separated from α s1 - and β -casein variants, as well as from whole casein, by DEAE-cellulose-urea chromatography. When κ -easein is in the reduced form it elutes from DEAE-cellulose at 0.05m NaCl.
