Recombination-assisted megaprimer (RAM) cloning
No molecular cloning technique is considered universally reliable, and many suffer from being too laborious, complex, or expensive. Restriction-free cloning is among the simplest, most rapid, and cost-effective methods, but does not always provide ...

In vivo cloning of up to 16 kb plasmids in E. coli is as simple as PCR. [PDF]
The precise assembly of defined DNA sequences into plasmids is an essential task in bioscience research. While a number of molecular cloning techniques have been developed, many methods require specialized expensive reagents or laborious experimental ...

Molecular cloning of schistosome genes
As part of an integrated programme investigating human schistosomiasis, work which involves epidemiological surveys and detailed immunological studies as well as biochemical investigations, we have, over the last three years, been cloning schistosome genes in a variety of plasmid and lambda vector systems.

The newfound relationship between extrachromosomal DNAs and excised signal circles
Extrachromosomal DNAs (ecDNAs) contribute to the progression of many human cancers. In addition, circular DNA by‐products of V(D)J recombination, excised signal circles (ESCs), have roles in cancer progression but have largely been overlooked. In this Review, we explore the roles of ecDNAs and ESCs in cancer development, and highlight why these ...

Molecular Cloning of Gonadotropin Hormones I (Gth-i) and II (Gth-ii) Genes in the Hard-lipped Barb (Osteochillus Hasseltii) and the Effect of Photoperiods on the Genes Expression [PDF]
Photoperiods is one of the factors that influence regulation on endocrine gland in producing hormones necessary for gonadal growth and development, gametogenesis and reproductive cycles in fish reproduction. This study clones the gonadotropin hormones I (

This study reveals how the mitochondrial protein Slm35 is regulated in Saccharomyces cerevisiae. The authors identify stress‐responsive DNA elements and two upstream open reading frames (uORFs) in the 5′ untranslated region of SLM35. One uORF restricts translation, and its mutation increases Slm35 protein levels and mitophagy.

Molecular cloning of the cDNA encoding pp36, a tyrosine-phosphorylated adaptor protein selectively expressed by T cells and natural killer cells. [PDF]
Activation of T and natural killer (NK) cells leads to the tyrosine phosphorylation of pp36 and to its association with several signaling molecules, including phospholipase Cgamma-1 and Grb2.

In situ molecular organization and heterogeneity of the Legionella Dot/Icm T4SS
We present a nearly complete in situ model of the Legionella Dot/Icm type IV secretion system, revealing its central secretion channel and identifying new components. Using cryo‐electron tomography with AI‐based modeling, our work highlights the structure, variability, and mechanism of this complex nanomachine, advancing understanding of bacterial ...
