Suppression of colorectal cancer liver metastasis by apolipoprotein(a) kringle V in a nude mouse model through the induction of apoptosis in tumor-associated endothelial cells.

open access: yesPLoS ONE, 2014
The formation of liver metastases in colorectal cancer patients is the primary cause of patient death. Current therapies directed at liver metastasis from colorectal cancer have had minimal impact on patient outcomes.

Genipin‐Crosslinked, Silane‐Anchored 3D Tumor–Stroma Microtissues for High‐Content On‐Chip Drug Testing

open access: yesAdvanced Healthcare Materials, EarlyView.
We describe a microfluidic tumor‐stroma co‐culture model, engineered to resist collagen‐hydrogel contraction driven by fibroblast activity. Surface silanization with APTES covalently anchors the matrix to the chip, while Genipin crosslinking progressively increases stiffness and elasticity without harming cells. This supports >10 days of co‐culture and

Liver metastases of colorectal cancer contain different subsets of tissue-resident memory CD8 T cells correlated with a distinct risk of relapse following surgery

open access: yesOncoImmunology
Tissue-resident memory (TRM) T cells have emerged as key players in cancer immunosurveillance, and their presence has been linked to a favorable clinical outcome in solid cancer patients.

One‐Step Microfluidic Manufactured Fucose‐Decorated Sweetosomes Choose the Time and the Road for Their Intracellular Journey to Cancer Treatment

open access: yesAdvanced Healthcare Materials, EarlyView.
Sweetosomes, a new class of fucose‐decorated liposomes, are developed via a one‐step microfluidic process without surface chemistry. This study elucidates their main caveolae‐mediated entry and distinct endosomal trafficking. These nanostructures demonstrate superior endosomal escape, organelle acidity modulation, and prolonged plasma persistence ...

Impact of multidisciplinary tumour boards (MTB) on the clinicopathological characteristics and outcomes of resected colorectal liver metastases across time

open access: yesWorld Journal of Surgical Oncology, 2020
Background Resection of colorectal liver metastases (CLM) has been established as the standard of care. This study aims to compare the change in clinicopathological characteristics of patients who underwent curative resection of CLM across two time ...

A Radioresistant‐Tumor‐Targeted Nanoparticle for X‐Ray‐Controlled Nitric Oxide Release to Potentiate Radiotherapy

open access: yesAdvanced Science, EarlyView.
The PBTN targets GRP78 and, upon X‐ray irradiation, generates ONOO− to induce immunogenic cell death, thereby activating CD8+ T cells and establishing a systemic antitumor immune response. ABSTRACT Nitric oxide (NO) treated radioresistant tumors by relieving hypoxia and blocking DNA repair, but its nonselective toxicity has precluded therapeutic use ...

SMAD4 Palmitoylation Drives a Metabolic‐Transcriptional Circuit to Promote Tumorigenesis and Confers Radiosensitivity in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
This study identifies palmitoylation as a novel regulatory modification of SMAD4, mediated by ZDHHC22/APT2. It activates fatty acid synthesis, creating a self‐reinforcing SMAD4–FASN–palmitate feedback loop that drives pancreatic cancer growth and enhances radiotherapy sensitivity.

Surgical treatment of colorectal cancer metastases [PDF]

open access: yesScripta Medica, 2020
Background/Aim: Colorectal metastatic liver tumours are the most common secondary liver tumours. During the life of patients with colorectal tumorous, this liver metastases will develop either synchronously or metachronously in half of the patients ...

Tumor‐Derived Alpha‐1 Antitrypsin Promotes Liver Metastasis of Colorectal Cancer Through the Neutrophil Extracellular Traps–CCDC25 Pathway

open access: yesAdvanced Science, EarlyView.
ABSTRACT Liver metastasis is a leading cause of mortality in colorectal cancer (CRC), where the inflammatory tumor microenvironment, specifically neutrophil infiltration, significantly promotes metastatic colonization. This study reveals a pro‐metastatic role for alpha‐1 antitrypsin (A1AT) in CRC liver metastasis via a dual mechanism involving ...

TSPYL5 Promotes Triple‐Negative Breast Cancer Metastasis by Antagonizing USP10‐Mediated PTEN Stabilization to Unleash a ZEB1‐Dependent EMT Program

open access: yesAdvanced Science, EarlyView.
The hyperactivation of PI3K/AKT signaling in PTEN wild‐type triple‐negative breast cancer represents a clinical paradox. We delineate a novel post‐translational regulatory axis wherein the oncogene TSPYL5 competitively antagonizes the deubiquitinase USP10.
