A Comparative Study: Flow Cytometry, Complement-dependent Cytotoxicity, and Luminex Methods

Transplantation Proceedings, 2019
The cell-based flow cytometric and bead-based Luminex crossmatch methods have been used alongside the standard complement-dependent cytotoxic crossmatch (CDCXM) test to detect donor specific anti-HLA antibodies. In this study, it was aimed to compare flow cytometric crossmatch (FCXM), CDCXM, and Luminex donor-specific crossmatch (LM-XM) tests for pre ...

Cancer resistance to complement-dependent cytotoxicity (CDC): Problem-oriented research and development

Molecular Immunology, 2009
The number of anti-cancer antibodies in therapy and in clinical trials is increasing gradually while their curative efficacy remains rather limited due to the resistance of tumor cells to complement-dependent cytotoxicity (CDC). An updated review of the various defense mechanisms complement is confronting when tackling a tumor cell is presented.

Mortalin inhibitors sensitize K562 leukemia cells to complement‐dependent cytotoxicity

International Journal of Cancer, 2009
AbstractMortalin, the mitochondrial hsp70, is a vital constitutively expressed heat shock protein. Its elevated expression has been correlated with malignant transformation and poor cancer prognosis. Cancer cells exhibit increased resistance to complement‐dependent cytotoxicity, partly due to their capacity to eliminate the complement membrane attack ...

A non-radioactive complement-dependent cytotoxicity assay for anti-CD20 monoclonal antibody

Journal of Immunological Methods, 1997
A simple and non-radioactive complement-dependent cytotoxicity assay was developed to determine the relative potency of an anti-CD20 mAb, IDEC-C2B8. The assay measures the relative number of viable cells based on the uptake and metabolism of the redox dye, Alamar blue.

Functional Analysis of CD59 Using Complement-Dependent Cytotoxicity Assay

2013
CD59 overexpression has been shown to confer the resistance of tumors to complement lysis. Complement lysis is one of the two major killing mechanisms of therapeutic anticancer antibodies. This chapter provides a method that allows studying the extent of complement protection of tumors by CD59.

Pretransplantation monitoring of HLA antibodies by complement dependent cytotoxicity and Luminex-based assays

Wiener klinische Wochenschrift, 2016
The introduction of more sensitive techniques, such as Luminex® for HLA antibody screening of patients awaiting organ transplantation has resulted in a better understanding of transplantation immunology and improvements in clinical practice. The interpretation of the results obtained only by Luminex® can lead to inaccurate evaluation of a patient's ...

Complement‐Dependent Cytotoxicity of Sera Obtained from Subacute Sclerosing Panencephalitis Patients

Microbiology and Immunology, 1978
AbstractHuman lymphoid cells (NC‐37) persistently infected with either measles virus (Schwarz and TYCSA strains) or subacute sclerosing panencephalitis (SSPE) virus (Halle and Mantooth strains) were destroyed in the presence of complement by anti‐measles sera as well as by sera from SSPE patients. The cytotoxic activity was demonstrated in both IgG and

COMPLEMENT‐DEPENDENT IN VITRO CYTOTOXICITY AGAINST AUTOLOGOUS INVASIVE BLADDER TUMOR CELLS IN HUMANS

Acta Pathologica Microbiologica Scandinavica Section C Immunology, 1981
Complement‐dependent serum‐mediated cytotoxicity (CDC) was measured in a 51‐chromium release assay against autologous tumor cells from 7 non‐invasive and 9 invasive transitional‐cell tumors of the urinary bladder. CDC was demonstrated against tumor cells from invasive tumors.

Conversion of IgG-mediated complement-dependent cytotoxicity by reducing reagents.

The Journal of heart and lung transplantation : the official publication of the International Society for Heart Transplantation, 1991
Reducing reagents used in complement-dependent crossmatches reduce IgM but not IgG. A positive IgM crossmatch is not an absolute contraindication to allografting. We have studied an alloantiserum from a patient with 100% panel reactive antibody awaiting heart transplantation whose serum was reduced.

The Selection of Aptamers to CD20 and Their Application as Inhibitors of Complement Dependent Cytotoxicity

2015
CD20 is an important oncological B-cell marker. Immunotherapy, using anti-CD20 antibodies, has revolutionized the treatment of B-cell cancers. Aptamers are highly specific DNA ligands, raised to identify virtually any target molecule through an iterative process known as SELEX (systematic evolution of ligands by exponential amplification).
