Epigenetics as a mechanism driving polygenic clinical drug resistance [PDF]

open access: yes, 2006
Aberrant methylation of CpG islands located at or near gene promoters is associated with inactivation of gene expression during tumour development.

Inhibition of SIRT7 Overcomes Radioresistance in Pancreatic Neuroendocrine Tumors by Reactivating MEN1 Expression

open access: yesAdvanced Science, EarlyView.
Pancreatic neuroendocrine tumors frequently silence MEN1 through epigenetic mechanisms. Here, SIRT7 recruits DNMT1 to the MEN1 promoter, drives hypermethylation, and enhances DNA repair. Inhibiting SIRT7 restores MEN1, reduces MRN complex abundance, impairs double‐strand break repair, and sensitizes PanNET models to radiation, supporting SIRT7 as a ...

CpG-C infiltrates the brain and is internalized by endothelial cells, astrocytes, and microglia.

open access: yes, 2019
(a-b) TAMRA-labeled CpG-C was injected intraperitoneally; 24 hours later, brains were perfused, and CpG-C internalization in endothelial cells (CD31), astrocytes (GFAP and GLAST), and microglia (CX3CR1 and CD11b) was visualized in histological sections ...

Metabolic Imbalance Triggers Adaptive Remodeling to Accelerate Diploidization in Murine Haploid Embryonic Stem Cells

open access: yesAdvanced Science, EarlyView.
In this article, Shuai and colleagues demonstrate that metabolic remodeling drives self‐diploidization in murine haploid ESCs (haESCs). Mitochondrial dysfunction and imbalanced pyruvate metabolism underlie this process. Genome‐wide screening using haESCs identifies key mitochondrial quality‐control related genes, enabling a metabolism‐based medium that

Recognition of 5-hydroxymethylcytosine by the Uhrf1 SRA domain. [PDF]

open access: yes, 2011
Recent discovery of 5-hydroxymethylcytosine (5hmC) in genomic DNA raises the question how this sixth base is recognized by cellular proteins. In contrast to the methyl-CpG binding domain (MBD) of MeCP2, we found that the SRA domain of Uhrf1, an essential

Epigenetic Silencing of RFX7 Defines a Transcriptional Axis Linking Lactate Metabolism to Immune Checkpoint Therapy in Glioblastoma

open access: yesAdvanced Science, EarlyView.
Glioblastoma (GBM) exhibits RFX7 silencing via promoter hypermethylation, which downregulates PIK3IP1 to activate PI3K/AKT signaling, increase lactate accumulation, and enhance histone H4K12 lactylation. This lactylation upregulates PD‐L1/CSF1 expression and drives the immunosuppressive microenvironment formation.

Impact of rock mineralogy on reactive transport of CO2 during carbon sequestration in a saline aquifer

open access: yesJournal of Petroleum Exploration and Production Technology
Deep saline aquifers offer a vast long-term storage capacity for CO2. The diverse mineral compositions in CO2 geological storage systems complicate the reactive transport of CO2.

Introgressed Variation in TaMYB7‐A1 Drives Graded Dormancy and Climate‐Adaptive Pre‐Harvest Sprouting Resistance in Wheat

open access: yesAdvanced Science, EarlyView.
TaMYB7‐A1 directly activates TaABI5 to enhance ABA signaling and regulate ABA‐GA homeostasis, enforcing seed dormancy. Its superior allele, derived from wild einkorn introgression, harbors a MITE insertion that elevates expression and two amino acid substitutions that enhance transcriptional activity, collectively generating graded PHS resistance for ...

Studies on CpG distribution in genomes

open access: yes, 2013
MS, DBTESDNA methylation is an epigenetic modification that plays very important role in vertebrate genomes. In vertebrate genomes, DNA methylation occurs at CpG sites and leads to gene regulation, gene imprinting, X-chromosome inactivation and cancer.
