ATF4‐mediated stress response as a therapeutic vulnerability in chordoma

open access: yesMolecular Oncology, EarlyView.
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

A finite element analysis of fatigue crack closure [PDF]

open access: yes
Experiments have shown that fatigue cracks close at positive loads during constant-amplitude load cycling. The crack-closure phenomenon is caused by residual plastic deformations remaining in the wake of an advancing crack tip.

Effects of Processing Residual Stresses on Fatigue Crack Growth Behavior of Structural Materials: Experimental Approaches and Microstructural Mechanisms [PDF]

open access: yes, 2012
Fatigue crack growth mechanisms of long cracks through fields with low and high residual stresses were investigated for a common structural aluminum alloy, 6061-T61.

Plecstatin inhibits hepatocellular carcinoma tumorigenesis and invasion through cytolinker plectin

open access: yesMolecular Oncology, EarlyView.
The ruthenium‐based metallodrug plecstatin exerts its anticancer effect in hepatocellular carcinoma (HCC) primarily through selective targeting of plectin. By disrupting plectin‐mediated cytoskeletal organization, plecstatin inhibits anchorage‐dependent growth, cell polarization, and tumor cell dissemination.

Therapeutic strategies for MMAE‐resistant bladder cancer through DPP4 inhibition

open access: yesMolecular Oncology, EarlyView.
We established monomethyl auristatin E (MMAE)‐resistant bladder cancer (BC) cell lines by exposure to progressively increasing concentrations of MMAE in vitro. RNA sequencing showed DPP4 expression was increased in MMAE‐resistant BC cells. Both si‐DPP4 and the DPP4 inhibitor sitagliptin suppressed the viability of MMAE‐resistant BC cells.

Fatigue crack growth in notched specimens: a numerical analysis

open access: yesFracture and Structural Integrity, 2019
Fatigue crack growth (FCG) is linked to irreversible and non-linear processes happening at the crack tip, which explains different problems observed in the use of da/dN-?K curves. The replacement of ?K by non-linear crack tip parameters, namely the crack

Crack Closure Effects on Fatigue Crack Propagation Rates: Application of a Proposed Theoretical Model

open access: yesAdvances in Materials Science and Engineering, 2016
Structural design taking into account fatigue damage requires a thorough knowledge of the behaviour of materials. In addition to the monotonic behaviour of the materials, it is also important to assess their cyclic response and fatigue crack propagation ...

Evaluation of the effect of crack closure on fatigue crack growth of simulated short cracks [PDF]

open access: yes
A test program was performed to determine the influence of crack closure on fatigue crack growth (FCG) rates of short cracks. By use of the standard compact tension specimen, test procedures were devised to evaluate closure loads in the wake of the crack

Dammarenediol II enhances etoposide‐induced apoptosis by targeting O‐GlcNAc transferase and Akt/GSK3β/mTOR signaling in liver cancer

open access: yesMolecular Oncology, EarlyView.
Etoposide induces DNA damage, activating p53‐dependent apoptosis via caspase‐3/7, which cleaves PARP1. Dammarenediol II enhances this apoptotic pathway by suppressing O‐GlcNAc transferase activity, further decreasing O‐GlcNAcylation. The reduction in O‐GlcNAc levels boosts p53‐driven apoptosis and influences the Akt/GSK3β/mTOR signaling pathway ...

Closure Effect of I + II Mixed-mode Crack for EA4T Axle Steel

open access: yesChinese Journal of Mechanical Engineering
The crack-closure effect is a crucial factor that affects the crack growth rate and should be considered in simulation analysis and testing. A mixed-mode I + II loading fatigue crack growth test was performed using EA4T axle steel specimens.
