A New Tool for CRISPR-Cas13a-Based Cancer Gene Therapy [PDF]

open access: yesMolecular Therapy - Oncolytics, 2020
Cas13a has already been successfully applied to virus detection. However, as a new gene interference tool, its potential in cancer treatment was not fully explored until now. This study constructed a new Cas13a expression vector, decoy minimal promoter-Cas13a-U6-guide RNA (DMP-Cas13a-U6-gRNA [DCUg]), by controlling the Cas13a and gRNA expression with a

Structure of AcrVIA2 and its binding mechanism to CRISPR-Cas13a

Biochemical and Biophysical Research Communications, 2022
Phages and non-phage derived bacteria have evolved many anti-CRISPR proteins (Acrs) to escape the adaptive immune system of prokaryotes. Thus Acrs can be applied as a regulatory tool for gene edition by CRISPR system. Recently, a non-phage derived AcrVIA2 has been identified as an inhibitor that blocks the editing activity of Cas13a in vitro by binding

RNA-mediated Allosteric Activation in CRISPR-Cas13a

2023
ABSTRACT Cas13a is a recent addition to the CRISPR-Cas toolkit that exclusively targets RNA, which makes it a promising tool for RNA detection. The protein uses a CRISPR RNA (crRNA) to target RNA sequences, which are cleaved by a composite active site formed by two ‘Higher Eukaryotes and Prokaryotes Nucleotide’ (HEPN) catalytic domains.

Rapid detection of Nipah virus using the one-pot RPA-CRISPR/Cas13a assay

open access: yesVirus Research, 2023
International audienceNipah virus (NiV) is a zoonotic pathogen with airborne transmission and high case fatality rates in humans. There is currently no treatment or vaccine against NiV infection approved for humans or animals, therefore early diagnosis ...

Rapid and facile detection of HBV with CRISPR/Cas13a

New Journal of Chemistry, 2022
A rapid point-of-care assay based on CRISPR-Cas13a for HBV was built with a LOD of 1 copy per μL was achieved in 50 min. The lateral flow test strip method can achieve naked-eye results, and the fluorescence readout can achieve real-time detection.

CRISPR–Cas13a: Prospects for Plant Virus Resistance

Trends in Biotechnology, 2018
CRISPR-Cas13a is an efficient RNA targeting and editing tool characterized recently in prokaryotes. This system can be recruited to engineer resistance against plant RNA viruses and regulate gene expression. We discuss some important achievements in using the CRISPR-Cas13a system to confer resistance against plant RNA viruses.

CRISPR/Cas13a-Based MicroRNA Detection in Tumor-Derived Extracellular Vesicles

open access: yesAdvanced Science, 2023
AbstractMicroRNAs (miRNAs) in extracellular vesicles (EVs) play essential roles in cancer initiation and progression. Quantitative measurements of EV miRNAs are critical for cancer diagnosis and longitudinal monitoring. Traditional PCR‐based methods, however, require multi‐step procedures and remain as bulk analysis.
