CRISPR–Cas12a with split crRNA for the direct and sensitive detection of microRNA

The Analyst
A split crRNA-activated Cas12a-based method enables amplification-free miRNA-21 detection. This method demonstrates enhanced sensitivity and specificity, achieving the accurate quantification of miRNA-21 in serum.

RNA-mediated CRISPR-Cas13 inhibition through crRNA structural mimicry

Science
To circumvent CRISPR-Cas immunity, phages express anti-CRISPR factors that inhibit the expression or activities of Cas proteins. Whereas most anti-CRISPRs described to date are proteins, recently described small RNAs called RNA anti-CRISPRs (rAcrs) have sequence homology to CRISPR RNAs (crRNAs) and displace them from cognate Cas nucleases.

Nitroreductase-Responsive Oligomeric crRNAs for Enzyme-Triggered Regulation of CRISPR Activity

The Journal of Organic Chemistry
Hypoxic tumors overexpress nitroreductase (NTR), providing an endogenous trigger for selective biomolecular activation. Here, we describe the synthesis of NTR-responsive clustered regularly interspaced short palindromic repeats (CRISPR) guide RNAs via the site-specific incorporation of a p-nitrobenzyl (p-NB) phosphoramidite at the 5' terminus of crRNAs.

Allosteric ribozyme-driven crRNA switch for the amplification-free detection of biomolecules

Biosensors and Bioelectronics
Currently, CRISPR-mediated biosensors are concentrating on the design of the crRNA or the activator strand to regulate the trans-cleavage activity of Cas12a. Herein, we report an allosteric ribozyme-driven crRNA switch-regulated CRISPR/Cas12a sensor for amplification-free detection of biomolecules.

Cryo-EM structure of the RfxCas13d-crRNA-off-target-RNA complex

Structure
The CRISPR-Cas system is crucial for the adaptive immune response of prokaryotes and has been widely applied for genetic engineering. Cas13d, a type VI-D CRISPR-Cas effector, functions as RNA-guided ribonuclease and has been engineered for programmable RNA editing, which is a commonly used, active, and well-characterized small type VI editor.
