Thinner is not always better: Optimizing cryo-lamellae for subtomogram averaging [PDF]

open access: yes
Cryo-electron tomography (cryo-ET) is a powerful method to elucidate subcellular architecture and to structurally analyze biomolecules in situ by subtomogram averaging, yet data quality critically depends on specimen thickness.

EM grid seeding for cryo-FIB milling v1

open access: yes
Defective mitochondrial quality control in response to loss of mitochondrial membrane polarization is implicated in Parkinson’s disease by mutations in PINK1 and PRKN. Application of in situ cryo-electron tomography (cryo-ET) made it possible to visualize the consequences of mitochondrial depolarization at higher resolution than heretofore attainable.

Whole cryo-FIB lamella acquisition at high magnification with rectangular beam montage cryo-electron tomography

open access: yesbioRxiv
The visualisation of large biological samples in cryo-electron tomography requires both few-nanometre resolution and a wide, ideally >10 µm, field of view.
