The D-type cyclins are among the candidate 'G1 cyclins' in higher eukaryotes that may regulate G1-S-phase progression. The human cyclin D1 gene, also known as PRAD1 (and previously as D11S287), is a putative proto-oncogene strongly implicated in several types of human tumors, including parathyroid adenomas, B-cell neoplasms (as the 'BCL-1 oncogene ...

Cell, 1991
A cDNA library prepared from a human glioblastoma cell line has been introduced into a budding yeast strain that lacks CLN1 and CLN2 and is conditionally deficient for CLN3 function. We rescued a gene that we call cyclin D1. It is related to A-, B-, and CLN-type cyclins, but appears to define a new subclass within the cyclin gene family.

Cyclin D as a therapeutic target in cancer
Nature Reviews Cancer, 2011Cyclin D1, and to a lesser extent the other D-type cyclins, is frequently deregulated in cancer and is a biomarker of cancer phenotype and disease progression. The ability of these cyclins to activate the cyclin-dependent kinases (CDKs) CDK4 and CDK6 is the most extensively documented mechanism for their oncogenic actions and provides an attractive ...

D-type cyclins couple extracellular signals to the biochemical machinery that governs progression through G1 phase of the mammalian cell division cycle. Induced by growth factor stimulation, D-type cyclins assemble with cyclin-dependent kinases CDK4 and CDK6 to form holoenzymes that facilitate exit from G1 by phosphorylating key substrates, including ...

The path to destruction for D-type cyclin proteins
Nature, 2021The molecular mechanism governing the destruction of key cell-cycle proteins, D-type cyclins, has been elucidated. This mechanism might underlie the lack of response of some human tumours to an inhibitor treatment. Mechanism discovered for the degradation of key cell-cycle regulators.

The D-type cyclins and their role in tumorigenesis
Journal of Cell Science, 1994SUMMARY The D-type cyclins are expressed during the progression from G0/G1 to S phase in the mammalian cell cycle. There is considerable evidence that they contribute to the development of specific cancers, both in humans and in mouse models.

Cyclin D expression in chronic lymphocytic leukemia
Leukemia & Lymphoma, 2005Although the majority of circulating leukemic cells in chronic lymphocytic leukemia (CLL) are in G0/early G1, recent studies have shown that these cells have undergone multiple cell divisions. In this study, we have determined whether there are abnormalities in cell cycle control in CLL by examining the three cyclin D isoforms in 43 patients and ...

Cyclin D-CDK4/6 functions in cancer
2020The mammalian cell cycle is driven by a complex of cyclins and their associated cyclin-dependent kinases (CDKs). Abnormal dysregulation of cyclin-CDK is a hallmark of cancer. D-type cyclins and their associated CDKs (CDK4 and CDK6) are key components of cell cycle machinery in driving G1 to S phase transition via phosphorylating and inactivating the ...

Hedgehog regulates cell growth and proliferation by inducing Cyclin D and Cyclin E
Nature, 2002Although mutations that activate the Hedgehog (Hh) signalling pathway have been linked to several types of cancer, the molecular and cellular basis of Hh's ability to induce tumour formation is not well understood. We identified a mutation in patched (ptc), an inhibitor of Hh signalling, in a genetic screen for regulators of the Retinoblastoma (Rb ...
