Psoralen, a mechanism-based inactivator of CYP2B6

Chemico-Biological Interactions, 2015
Furanocoumarin compound psoralen (PRN) is a major active ingredient found in herbaceous plants. PRN has been used for the treatment of various dermal diseases in China. We evaluated the inhibitory effect of PRN on cytochrome P450 2B6 (CYP2B6) and found that PRN induced a time-, concentration-, and NADPH-dependent inactivation of CYP2B6 with the values ...

Imperatorin Is a Mechanism-Based Inactivator of CYP2B6

Drug Metabolism and Disposition, 2015
Imperatorin (IMP) is the major active ingredient in many common medicinal herbs. We examined the irreversible inhibitory effect of IMP on CYP2B6. IMP produced a time- and concentration-dependent inactivation of CYP2B6. About 70% of activity of CYP2B6 was suppressed after its incubation with 1.5 μM IMP for 9 minutes. KI and kinact were found to be 0.498

Circulating miRNAs as Biomarkers for CYP2B6 Enzyme Activity

Clinical Pharmacology & Therapeutics, 2020
The CYP2B6 gene is highly polymorphic and its activity shows wide interindividual variability. However, substantial variability in CYP2B6 activity remains unexplained by the known CYP2B6 genetic variations. Circulating, cell‐free micro RNAs (miRNAs) may serve as biomarkers of hepatic enzyme activity.

[Cyclophosphamide and CYP2B6].

Gan to kagaku ryoho. Cancer & chemotherapy, 2008
Members of the hepatic cytochrome p450 superfamily(CYP)play an important role in the metabolism of many therapeutic drugs. There is a marked individual variability in the CYP-mediated drug metabolism. One of the main factors of the CYP activities is a polymorphism of the CYP genes.

Cytochrome P450 CYP2B6 genotypes and haplotypes in a Colombian population

Pharmacogenetics and Genomics, 2011
Information on CYP2B6 allele frequencies and detrimental genotypes in mixed human populations is scarce. The aim of this study was to analyze the frequencies and haplotypes of nonsynonymous CYP2B6 single nucleotide polymorphisms (SNPs) in a Colombian population. One hundred and fifty-two healthy individuals were analyzed for five nonsynonymous CYP2B6 ...

Contribution of CYP2B6 alleles in explaining extreme (S)-methadone plasma levels

Pharmacogenetics and Genomics, 2013
BACKGROUND: (S)-Methadone, metabolized mainly by CYP2B6, shows a wide interindividual variability in its pharmacokinetics and pharmacodynamics. METHODS: Resequencing of the CYP2B6 gene was performed in 12 and 35 selected individuals with high (S)-methadone plasma exposure and low (S)-methadone plasma exposure, respectively, from a previously described ...

Detection of single nucleotide polymorphisms in CYP2B6 gene

2002
Publisher Summary Studies with new antibodies of higher sensitivity and specificity have shown that CYP2B6 is expressed in the liver of most or all individuals, but at extremely different levels, ranging from nearly undetectable up to about 80 pmol/mg of microsomal protein, which corresponds to about one-fifth of the total P450 content of the liver ...

Hepatic CYP2B6 Expression: Gender and Ethnic Differences and Relationship to CYP2B6 Genotype and CAR (Constitutive Androstane Receptor) Expression

The Journal of Pharmacology and Experimental Therapeutics, 2003
CYP2B6 metabolizes many drugs, and its expression varies greatly. CYP2B6 genotype-phenotype associations were determined using human livers that were biochemically phenotyped for CYP2B6 (mRNA, protein, and CYP2B6 activity), and genotyped for CYP2B6 coding and 5'-flanking regions. CYP2B6 expression differed significantly between sexes.

Activity Variations of CYP2B6 Determine the Metabolic Stratification of Efavirenz

Chemical Research in Toxicology
To investigate the effects of hepatic enzyme activity variations and CYP2B6 gene polymorphisms on the in vivo and in vitro metabolism of efavirenz. In vitro enzyme systems using rat and human liver microsomes (RLM/HLM) were established, with in vivo studies conducted on Sprague-Dawley rats. Metabolite detection was performed via LC-MS/MS.
