Involvement of CYP2D6 and CYP2B6 on Tramadol Pharmacokinetics
Pharmacogenomics, 2020This study included 24 healthy volunteers who received a single 37.5 mg oral dose of tramadol. We analyzed 18 polymorphisms within CYP2D6, CYP2B6, CYP3A, COMT, ABCB1, SLC22A1 and OPRM1 genes by quantitative PCR, to study whether these polymorphisms affect its pharmacokinetics, pharmacodynamics and safety. CYP2D6 intermediate metabolizers (n = 6) showed

CYP2B6 metabolizes many drugs, and its expression varies greatly. CYP2B6 genotype-phenotype associations were determined using human livers that were biochemically phenotyped for CYP2B6 (mRNA, protein, and CYP2B6 activity), and genotyped for CYP2B6 coding and 5'-flanking regions. CYP2B6 expression differed significantly between sexes.

Human Liver CYP2B6-Catalyzed Hydroxylation of RP 73401
The Journal of Pharmacology and Experimental Therapeutics, 1997RP 73401 is a potent inhibitor of cyclic nucleotide phosphodiesterase type IV. RP 73401 is metabolized by human liver microsomes almost exclusively by transhydroxylation of the cyclopentyl group to RPR 113406. Liquid chromatography/mass spectrometry/mass spectrometry analysis of plasma from patients given RP 73401 also revealed a molecular ion and ...

Circulating miRNAs as Biomarkers for CYP2B6 Enzyme Activity
Clinical Pharmacology & Therapeutics, 2020The CYP2B6 gene is highly polymorphic and its activity shows wide interindividual variability. However, substantial variability in CYP2B6 activity remains unexplained by the known CYP2B6 genetic variations. Circulating, cell‐free micro RNAs (miRNAs) may serve as biomarkers of hepatic enzyme activity.

Efavirenz Intoxication Due to a New Cyp2B6 Constellation
Antiviral Therapy, 2013Here, we describe severe neuropsychiatric symptoms in an HIV-positive Asian man with extremely high efavirenz plasma levels while receiving standard treatment with efavirenz/tenofovir/emtricitabine fixed-dose regimen. Genetic examination revealed compound homozygosity for loss-of-function alleles of CYP2B6, including coding for a rare truncated ...

Cytochrome P450 CYP2B6 genotypes and haplotypes in a Colombian population
Pharmacogenetics and Genomics, 2011Information on CYP2B6 allele frequencies and detrimental genotypes in mixed human populations is scarce. The aim of this study was to analyze the frequencies and haplotypes of nonsynonymous CYP2B6 single nucleotide polymorphisms (SNPs) in a Colombian population. One hundred and fifty-two healthy individuals were analyzed for five nonsynonymous CYP2B6 ...

[Cyclophosphamide and CYP2B6].
Gan to kagaku ryoho. Cancer & chemotherapy, 2008Members of the hepatic cytochrome p450 superfamily(CYP)play an important role in the metabolism of many therapeutic drugs. There is a marked individual variability in the CYP-mediated drug metabolism. One of the main factors of the CYP activities is a polymorphism of the CYP genes.

THE ROLE OF CYP2B6 IN HUMAN XENOBIOTIC METABOLISM*
Drug Metabolism Reviews, 1999The cytochromes P450 (CYP) comprise a large multigene family of heme-thiolate proteins which are of considerable importance in the metabolism of xenobiotics and endobiotics.

CYP2B6 gene single nucleotide polymorphisms and leukemia susceptibility
Annals of Hematology, 2010CYP2B6 is a highly variable and polymorphic cytochrome P450 enzyme which plays a vital role in the degradation of some endogenous metabolites, xenobiotics, and harmful compounds. The 516G>T single nucleotide polymorphism (SNP) in exon 4 of CYP2B6 gene may change CYP2B6 enzyme activity and the gene expression in the liver.

Imperatorin Is a Mechanism-Based Inactivator of CYP2B6
Drug Metabolism and Disposition, 2015Imperatorin (IMP) is the major active ingredient in many common medicinal herbs. We examined the irreversible inhibitory effect of IMP on CYP2B6. IMP produced a time- and concentration-dependent inactivation of CYP2B6. About 70% of activity of CYP2B6 was suppressed after its incubation with 1.5 μM IMP for 9 minutes. KI and kinact were found to be 0.498
