Understanding CYP2D6 interactions
Drug Discovery Today, 2009Owing to the polymorphic nature of CYP2D6, clinically significant issues can arise when drugs rely on that enzyme either for clearance, or metabolism to an active metabolite. Available screening methods to determine if the compound is likely to cause drug-drug interactions, or is likely to be a victim of inhibition of CYP2D6 by other compounds will be ...

Drug Metabolism and Disposition, 2006
Unexplained cases of CYP2D6 genotype/phenotype discordance continue to be discovered. In previous studies, several African Americans with a poor metabolizer phenotype carried the reduced function CYP2D6*10 allele in combination with a nonfunctional allele.

Drug Metabolism and Pharmacokinetics, 2002
Novel single nucleotide polymorphisms (SNPs) were found in introns 1, 3, 4, 5, and 7 of the gene encoding cytochrome (CYP) 2D6 in 90 Japanese subjects. Some of these SNPs were associated with C2850T and/or C100T, which are key SNPs for CYP2D6(*)2 and CYP2D6(*)10, respectively.

Induction of CYP2D6 in pregnancy*
Clinical Pharmacology & Therapeutics, 1997Expression of the drug-metabolizing enzyme cytochrome P4502D6 (CYP2D6) is predominantly under genetic control, and enzyme-inducing drugs have little influence on its activity. We studied the activity of CYP2D6 during pregnancy. One hundred forty pregnant women were genotyped for CYP2D6.

Publisher Summary This chapter describes methods for the detection of the main CYP2D6 alleles associated with the PM phenotype and discusses the use of single-strand conformational polymorphism (SSCP) analysis and DNA sequencing for the identification of new polymorphisms. The cytochrome P450 enzyme debrisoquine 4-hydroxylase, which is encoded by the

Altered expression of CYP2D6 (debrisoquine hydroxylase), resulting from genetic polymorphism at the CYP2D6 gene locus, is responsible for pronounced interindividual variation in the metabolism of many clinically important drugs. Although CYP2D6 substrates are structurally diverse, most are small molecules that interact with the protein via an ...
