Synthesis of cysteine proteinase inhibitors structurally based on the proteinase interacting N-terminal region of human cystatin C

open access: yes, 1990
Peptides spanning the entire, or part of, the Gly4-Glu21 segment of the human cysteine proteinase inhibitor cystatin C have been synthesized. Peptides containing residues on the N-terminal side of Gly11 were rapidly cleaved by papain at the bond Gly11 ...

Acid‐Responsive Nanobot‐Integrated Core‐Shell Microneedles Reprogram the Degenerative Annulus Fibrosus Microenvironment Through Epigenetic Suppression of Ferroptosis

open access: yesAdvanced Science, EarlyView.
An acid‐responsive core‐shell microneedle system normalizes the degenerative annulus fibrosus microenvironment by consuming pathological H+, generating O2, and providing NIR‐amplified redox buffering. Mechanistically, it epigenetically suppresses ferroptosis via DNMT3A‐dependent methylation of the ATF3 promoter, relieving ATF3‐mediated repression of ...

Decoding the role of cysteine metabolism in angiogenesis

open access: yes
reservedAngiogenesis is the process by which new blood vessels are formed from pre existing ones in response to signals triggered by pathophysiological conditions, such as wound healing or cancer. In this study, we aim to decode the role of cysteine in

Asymmetric Structure‐Driven Functional Synergy: A Multistimuli‐Activated Janus Nanomotor for Self‐Amplifying NO Cascade Therapy

open access: yesAdvanced Science, EarlyView.
The Janus nanomotors leverage an “asymmetric structure‐driven functional synergy” strategy to orchestrate spatiotemporally coordinated catalytic functions, in situ self‐amplifying NO generation, and deep tumor penetration, thereby enabling controllable, sustained, and uniform NO generation within the tumor microenvironment.

A Hepatocyte‐to‐Stellate Cell Axis Couples Alternate‐Day Fasting to Liver Fibrosis Resolution via ATG7 S‐Nitrosylation

open access: yesAdvanced Science, EarlyView.
Alternate‐day fasting suppresses mitochondrial complex II assembly via SDHAF4 to upregulate hepatocyte eNOS. The resulting nitric oxide S‐nitrosylates ATG7 at C184 in hepatic stellate cells, restraining autophagic flux and preventing their profibrotic activation.

The Deubiquitinase UCHL1 Drives Susceptibility to Atrial Fibrillation by Stabilizing CaMKII‐δ

open access: yesAdvanced Science, EarlyView.
UCHL1 is highly upregulated in mice and patients with AF. UCHL1 directly binds to and removes K48‐linked polyubiquitin chains from CaMKII‐δ at K251 and enhances CaMKII‐δ stability, which activates RyR2, disrupts Ca2+ handling, and promotes AF. These findings establish UCHL1 as a critical regulator of CaMKII‐δ stability and AF‐related Ca2+ handling ...

Persistent Zn2+ Influx‐Mediated Deacetylation of ANXA2 Regulates the “Zn2+‐Autophagic Flux” and Alleviates Intervertebral Disc Degeneration

open access: yesAdvanced Science, EarlyView.
Zinc transporter SLC39A14 sustains intracellular Zn2+ levels to activate SIRT3‑dependent ANXA2 deacetylation at K302. This modification weakens ANXA2‑mTOR binding and releases autophagic‑flux suppression in nucleus pulposus cells. Dysregulated Zn2+ homeostasis disrupts this axis to drive intervertebral disc degeneration.
