Cytoplasmic p21 promotes stemness of colon cancer cells via activation of the NFκB pathway

open access: yesMolecular Oncology, EarlyView.
Cytoplasmic p21 promotes colorectal cancer stem cell (CSC) features by destabilizing the NFκB–IκB complex, activating NFκB signaling, and upregulating BCL‐xL and COX2. In contrast to nuclear p21, cytoplasmic p21 enhances spheroid formation and stemness transcription factor CD133.

Using regression trees for improving the efficiency of nonlinear programming solvers

open access: yesEngineering Science and Technology, an International Journal
Optimization plays a critical role in fields such as economics, engineering, and computational sciences, where finding the optimal values of decision variables is essential for the design of a product, production system, or service system.

In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Dual targeting of RET and SRC synergizes in RET fusion‐positive cancer cells

open access: yesMolecular Oncology, EarlyView.
Despite the strong activity of selective RET tyrosine kinase inhibitors (TKIs), resistance of RET fusion‐positive (RET+) lung cancer and thyroid cancer frequently occurs and is mainly driven by RET‐independent bypass mechanisms. Son et al. show that SRC TKIs significantly inhibit PAK and AKT survival signaling and enhance the efficacy of RET TKIs in ...

Methylation biomarkers can distinguish pleural mesothelioma from healthy pleura and other pleural pathologies

open access: yesMolecular Oncology, EarlyView.
We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.

unified approach to the construction of higher-order derivative-free iterative methods for solving systems of nonlinear equations

open access: yesProceedings of the Mongolian Academy of Sciences
In this article, we introduce a unified approach to constructing a higher-order derivative-free scheme based on the approximations of F'(zk)-1. A family of order  p=6,7 derivative-free method is proposed and compared to some well-known methods.

Detection of circulating tumor DNA in colorectal cancer patients using a methylation‐specific droplet digital PCR multiplex

open access: yesMolecular Oncology, EarlyView.
We developed a cost‐effective methylation‐specific droplet digital PCR multiplex assay containing tissue‐conserved and tumor‐specific methylation markers. The assay can detect circulating tumor DNA with high accuracy in patients with localized and metastatic colorectal cancer.

Impact of fractional and integer order derivatives on the (4+1)-dimensional fractional Davey–Stewartson–Kadomtsev–Petviashvili equation

open access: yesPartial Differential Equations in Applied Mathematics
In this study, the closed-form wave solutions of the (4+1)-dimensional fractional Davey–Stewartson–Kadomtsev–Petviashvili equation are investigated using the modified auxiliary equation method and the Jacobi elliptic function method. In the analysis, two

Perspectives in educating molecular pathologists on liquid biopsy: Toward integrative, equitable, and decentralized precision oncology

open access: yesMolecular Oncology, EarlyView.
Liquid biopsy enables minimally invasive, real‐time molecular profiling through analysis of circulating biomarkers in biological fluids. This Perspective highlights the importance of training pathologists through integrative educational programs, such as the European Masters in Molecular Pathology, to ensure effective and equitable implementation of ...

A partially derivative-free cyclic block coordinate descent method for nonseparable composite optimization

open access: yesMathematical Modelling and Analysis
In this paper, we address a composite optimization problem in which the objective function consists of two terms: the first presents a function with a gradient that satisfies a Lipschitz–Hölder composition, while the second one is a convex function ...
