Advanced Science, EarlyView.A liquid‐phase ion trap (LPIT) coupled with ESI‐MS enables rapid, high‐fidelity de novo sequencing of long oligonucleotides in ∼30 min. By applying an electric field to dissociate metal adducts in solution, LPIT delivers clean spectra, superior mass accuracy, and full sequence coverage, surpassing MALDI‐TOF and LC‐MS.
