Method development and validation of an analytical quality by design ultrafast liquid chromatographic method for the determination of bedaquiline from pharmaceutical bulk and nanoemulsions

open access: yesBiomedical Chromatography, Volume 38, Issue 12, December 2024.
Abstract Bedaquiline (BDQ) is a drug used to treat multidrug‐resistant tuberculosis (MDR‐TB). It exhibits exposure‐dependent efficacy in eliminating Mycobacterium tuberculosis (Mtb). An easy, efficient and precise reverse‐phase ultrafast liquid chromatography (RP‐UFLC) method was developed to validate the free base of the antitubercular medication BDQ.

TBAJ-587, a novel diarylquinoline, is active against Mycobacterium abscessus

open access: yesAntimicrobial Agents and Chemotherapy
ABSTRACT Nontuberculous mycobacteria (NTM) infections are extremely difficult to treat due to a natural resistance to many antimicrobials. TBAJ-587 is a novel diarylquinoline, which shows higher anti-tuberculosis activity, lower lipophilicity, and weaker inhibition of hERG channels than bedaquiline ...

Exploring the Chemical Space of Mycobacterial Oxidative Phosphorylation Inhibitors Using Molecular Modeling

open access: yesChemMedChem, Volume 19, Issue 22, November 18, 2024.
Intracellular pathogens are exceedingly clinically challenging, including pathogenic mycobacteria. Many global efforts are currently in action to battle these notorious disease agents. This review provides an overview of mycobacteria, along with their associated diseases, pathogenesis, and exploitable respiratory vulnerabilities to supplement ...

Clinical efficacy and computed tomography diagnostic value of bedaquiline‐containing regimens in the treatment of drug‐resistant pulmonary tuberculosis

open access: yesiLABMED, Volume 2, Issue 3, Page 149-156, September 2024.
This paper emphasizes the key role of Bedazoline Tablets in the treatment of drug‐resistant tuberculosis in turning negative sputum culture and improving absorption of chest CT lesions. It is helpful to improve the cure rate of drug‐resistant tuberculosis patients.

The Diarylquinoline R207910 Is Bactericidal against Mycobacterium leprae in Mice at Low Dose and Administered Intermittently [PDF]

open access: yesAntimicrobial Agents and Chemotherapy, 2009
ABSTRACT The diarylquinoline R207910 is profoundly bactericidal in a murine model of tuberculosis. Previously, R207910 was also found to be bactericidal for Mycobacterium leprae -infected mice during lag phase.

Subcutaneous infection caused by Mycobacterium abscessus following botulinum toxin injections: A case report and literature review

open access: yesJournal of Cosmetic Dermatology, Volume 23, Issue 5, Page 1527-1532, May 2024.
Abstract Background The rapid development of cosmetic injections has led to an increased incidence of nontuberculous mycobacterial (NTM) infection. Patients and Methods Here, we presented a case of cutaneous Mycobacterium abscessus infection subsequent to botulinum toxin injection for treating masseter hypertrophy, and reviewed the literature on skin ...

Safety, pharmacokinetics, and food effect of sudapyridine (WX‐081), a novel anti‐tuberculosis candidate in healthy Chinese subjects

open access: yesClinical and Translational Science, Volume 17, Issue 2, February 2024.
Abstract This study aimed to assess the safety, pharmacokinetics, and food impact on sudapyridine (WX‐081), a novel drug designed to inhibit mycobacterium ATP synthase, with clinical applications for drug‐resistant tuberculosis (TB) treatment. The research comprised two arms: a single ascending dose (SAD) arm (30 to 600 mg, N = 52) and a multiple ...

Diarylquinoline TMC207 as potential new therapy for multidrug-resistant tuberculosis [PDF]

open access: yesThorax, 2009
TMC207 is an investigational diarylquinoline which inhibits mycobacterial ATP synthase in drug-sensitive and drug-resistant Mycobacterium tuberculosis in vitro. It is bactericidal to dormant tubercle bacilli, therefore offering a new mechanism for the treatment of multidrug-resistant tuberculosis.

Mycobacterium tuberculosis: immune response, biomarkers, and therapeutic intervention

open access: yesMedComm, Volume 5, Issue 1, January 2024.
Currently, controlling the source of infection, interrupting transmission pathways, and protecting susceptible populations are key measures in tackling MTB infection. However, preventative, diagnostic, and therapeutic interventions for TB are also important in reducing the burden of the disease. First, in terms of preventing MTB infection, the Bacillus
