Roles of Ferric Peroxide Anion Intermediates (Fe3+O2−, Compound 0) in Cytochrome P450 19A1 Steroid Aromatization and a Cytochrome P450 2B4 Secosteroid Oxidation Model

open access: yesAngewandte Chemie, Volume 136, Issue 33, August 12, 2024.
Although most cytochrome P450‐catalyzed oxidations are believed to utilize perferryl oxygen (Compound I) as a reactive iron species, the ferric peroxide species (Compound 0) is thought to catalyze some oxidative deformylations. This study revealed the dominant contribution of Compound 0 in the P450 19A1‐catalyzed steroid aromatization and P450 2B4 ...

Recent developments in fluorine‐containing pesticides

open access: yesPest Management Science, Volume 80, Issue 7, Page 3065-3087, July 2024.
In the timeframe 2016–2022, approximately 77% of the 39 launched products (herbicides, fungicides, insecticides, acaricides, and nematicides) were halogen‐substituted and from these, approximately 64% were fluorine‐containing pesticides. Abstract To ensure ongoing sustainability, the modern agrochemical industry is faced with enormous challenges. These

Cholesterol-Dependent Degradation of Squalene Monooxygenase, a Control Point in Cholesterol Synthesis beyond HMG-CoA Reductase [PDF]

open access: yes, 2011
SummaryExquisite control of cholesterol synthesis is crucial for maintaining homeostasis of this vital yet potentially toxic lipid. Squalene monooxygenase (SM) catalyzes the first oxygenation step in cholesterol synthesis, acting on squalene before ...

Synthesis of 15.ALPHA.-Fluoro-24,25-dihydrolanosterol as a Potential Inhibitor and/or Mechanistic Probe for Lanosterol 14.ALPHA.-Demethylase.

open access: yesChemical and Pharmaceutical Bulletin, 2000
As a potential inhibitor and/or mechanistic probe for lanosterol 14alpha-demethylase, 15alpha-fluoro-24,25-dihydrolanosterol was prepared by fluorination of 15alpha-hydroxy-24,25-dihydrolanost-7-en-3beta-yl benzoate with diethylaminosulfur trifluoride, followed by hydrogen chloride-catalyzed isomerization of the delta7 to delta8 and reductive cleavage ...

7-Oxo-24,25-dihydrolanosterol: a novel lanosterol 14α-demethylase (P-45014) inhibitor which blocks electron transfer to the oxyferro intermediate [PDF]

open access: yesBiochimica et Biophysica Acta (BBA) - Lipids and Lipid Metabolism, 1987
7-Oxo-24,25-dihydrolanosterol (3 beta-hydroxy-8-lanosten-7-one, 7-oxo-HDL) was a potent competitive inhibitor for lanosterol 14 alpha-demethylase (cytochrome P-45014DM) of Saccharomyces cerevisiae. Affinity of 7-oxo-DHL for the enzyme was more than 50-times higher than those of the inherent substrates, lanosterol and 24,25-dihydrolanosterol.

Perturbation of sterol biosynthesis by itraconazole and ketoconazole in Leishmania mexicana mexicana infected macrophages.

open access: yes, 1989
The azole antifungals ketoconazole and itraconazole possess in vitro antileishmanial activity against Leishmania mexicana mexicana amastigotes in macrophages (cell line J774G8). As in yeast and fungi, the activity is likely to be due to inhibition of the
