vic-Diol chirons: Enantiospecific synthesis of 11,12- and 14,15-dihydroxyeicosatrienoic acids

Tetrahedron Letters, 1994
Abstract A differentiated, six-carbon vicinal -diol chiron was prepared from D -glucurono-6,3-lactone and exploited in the asymmetric synthesis of the erythro/threo -isomers of the title eicosanoids.

Effects of dronedarone, amiodarone and their active metabolites on sequential metabolism of arachidonic acid to epoxyeicosatrienoic and dihydroxyeicosatrienoic acids

Biochemical Pharmacology, 2017
Cardiac enzymes such as cytochrome P450 2J2 (CYP2J2) metabolize arachidonic acid (AA) to cardioprotective epoxyeicosatrienoic acids (EETs), which in turn are metabolized by soluble epoxide hydrolase (sEH) to dihydroxyeicosatrienoic acids (DHETs). As EETs and less potent DHETs exhibit cardioprotective and vasoprotective functions, optimum levels of ...

Dihydroxyeicosatrienoic Acid, a Metabolite of Epoxyeicosatrienoic Acids Upregulates Endothelial Nitric Oxide Synthase Expression Through Transcription: Mechanism of Vascular Endothelial Function Protection

Cell Biochemistry and Biophysics, 2021
The present study aimed to investigate the impacts and underlying mechanisms of 14,15-DHETs on eNOS and vascular endothelial functions. Bovine aortic endothelial cells (BAECs) were treated with various concentrations of 14, 15-DHET. The expressions of eNOS protein and mRNA were observed at different time points.

14,15-Dihydroxyeicosatrienoic acid relaxes bovine coronary arteries by activation of KCachannels

American Journal of Physiology-Heart and Circulatory Physiology, 2002
Epoxyeicosatrienoic acids (EETs) cause vascular relaxation by activating smooth muscle large conductance Ca2+-activated K+(KCa) channels. EETs are metabolized to dihydroxyeicosatrienoic acids (DHETs) by epoxide hydrolase. We examined the contribution of 14,15-DHET to 14,15-EET-induced relaxations and characterized its mechanism of action.

Total synthesis of 11-R,12-R-dihydroxyeicosatrienoic acid, a metabolite of the cytochrome P-450 epoxygenase pathway

Tetrahedron Letters, 1994
The first total and enantioselective synthesis of 11-R,12-R-dihydroxy-5,8,14-eicosatrienoic acid 7 is reported. We have used the carbohydrate 2-deoxy-D-glucose as a masked 1,6-dialdehyde precursor 20 to perform the synthesis. The availability of this material will allow, among other things, the testing of the hypothesis that it can be a biochemical ...

A sensitive LC-MS/MS method for the quantification of regioisomers of epoxyeicosatrienoic and dihydroxyeicosatrienoic acids in human plasma during endothelial stimulation

Analytical and Bioanalytical Chemistry, 2016
Epoxyeicosatrienoic acids (EETs) are vasodilating lipid mediators metabolized into dihydroxyeicosatrienoic acids (DHETs) by soluble epoxide hydrolase. We aimed to develop a LC-MS/MS method to quantify EETs and DHETs in human plasma and monitored their levels during vascular endothelial stimulation.

Biosynthesis of 5,6-dihydroxyprostaglandin E1, and F1α from 5,6-dihydroxyeicosatrienoic acid by ram seminal vesicles

Biochimica et Biophysica Acta (BBA) - Lipids and Lipid Metabolism, 1984
cis-5(6)Epoxy-cis-8,11,14-eicosatrienoic acid was recently found to be metabolized by ram seminal vesicles to 5-hydroxyprostaglandin I 1 alpha and 5-hydroxyprostaglandin I 1 beta, 5(6)epoxyprostaglandin E1 and 5,6-dihydroxyprostaglandin E1. The epoxide can be hydrolyzed by epoxide hydrolases to 5,6-dihydroxy-8,11,14-eicosatrienoic acid.
