Esters of okadaic acid and dinophysistoxin-2 in Portuguese bivalves related to human poisonings

Liquid chromatography (HPLC) was used to search for esters of DSP toxins in Portuguese bivalves. Hexane-soluble derivatives of okadaic acid (OA) and dinophysistoxin-2 (DTX-2) were found. Presumably they are acyl derivatives, globally known as 'dinophysistoxin-3' (DTX-3). In certain instances DTX-3 content surpassed 50% of the total amount of DSP toxins.

Abstract The production of diarrhetic shellfish poisoning toxins (okadaic acid analogues and other lipophilic toxins) by a culture of Dinophysis acuminata, fed with the autotrophic ciliate Myrionecta rubra, was confirmed by LC–MS analysis, and the toxin profile compared with that in the field assemblage of the same species.

Liquid chromatographic determination of okadaic acid and dinophysistoxin-1 in shellfish after derivatization with 9-chloromethylanthracene

The reagent 9-chloromethylanthracene was evaluated for derivatization of the diarrhetic shellfish poisons, okadaic acid and dinophysistoxin-1 (DTX-1), to form fluorescent products separable by liquid chromatography. The toxins were reacted with the reagent in acetonitrile in the presence of tetramethylammonium hydroxide for 1 h at 90 degrees C.

Different dinophysistoxin's profiles have been found repeatedly amongst some offshore bivalve molluscs. Species such as the clam, Spisula solida, esterify dinophysistoxins to a great extent, and contain always more okadaic acid (OA) than dinophysistoxin-2 (DTX2).
