Metabolic engineering of the diosgenin biosynthesis pathway in Trigonella foenum-graceum hairy root cultures

Plant Science, 2022
Diosgenin as a triterpene with numbers of pharmaceutical applications has been identified in Trigonella foenum-graceum. In this survey, in order to scale up the amount of diosgenin in Fenugreek as a promising alternative of yam, ∆24-reductase as a rate limiting enzyme in diosgenin biosynthesis pathway has been overexpressed by utilizing pBI121 ...

22R‐ but not 22S‐hydroxycholesterol is recruited for diosgenin biosynthesis

The Plant Journal, 2021
SUMMARYDiosgenin is an important compound in the pharmaceutical industry and it is biosynthesized in several eudicot and monocot species, herein represented by fenugreek (a eudicot), and Dioscorea zingiberensis (a monocot). Formation of diosgenin can be achieved by the early C22,16‐oxidations of cholesterol followed by a late C26‐oxidation.

Biosynthesis of diosgenin in Dioscorea tubers

Archives of Biochemistry and Biophysics, 1961
Abstract The incorporation of radioacetate into diosgenin by homogenates of Dioscorea floribunda has been demonstrated by rigorous purification of the isolated sapogenin. Mevalonic acid is not incorporated into diosgenin to any significant extent under these conditions.

Deciphering the network of cholesterol biosynthesis in Paris polyphylla laid a base for efficient diosgenin production in plant chassis

Metabolic Engineering, 2023
Cholesterol serves as a key precursor for many high-value chemicals such as plant-derived steroidal saponins and steroidal alkaloids, but a plant chassis for effective biosynthesis of high levels of cholesterol has not been established. Plant chassis have significant advantages over microbial chassis in terms of membrane protein expression, precursor ...

Kinetics of cellular growth and diosgenin biosynthesis in Dioscorea caucasica cell suspensions, cultured in batch system

Plant Science, 1992
Abstract By selecting small cell aggregates, several cell lines of Dioscorea with different characteristics were established: colour, size of cell aggregates, growth, accumulation of carotenoids, diosgenin and phytosterols. The white-coloured lines accumulate larger amounts of diosgenin than the unselected cell suspension and the cream-coloured ...

Expression analysis of steroid pathway genes revealed positive correlation with diosgenin biosynthesis in Trillium govanianum

Acta Physiologiae Plantarum, 2016
Trillium govanianum Wall. ex D. Don, popularly known as Nagchhatri, is an important medicinal plant of northwestern Himalayas. No data exist on molecular characteristics of diosgenin biosynthesis, the main chemical component of T. govanianum. HPLC analysis revealed diosgenin content of 2.4 and 0.7% in hydrolyzed rhizome extracts of Chamba and Lahaul ...
