An Oligomeric Ser-Pro Dipeptide Mimetic Assuming the Polyproline II Helix Conformation

A hexapeptide assembled from dipeptide building blocks assumes the secondary structure of the polyproline II (PPII) helix. Side chain-to-backbone hydrogen bonds stabilize peptide torsions next to the fused ring systems. The solution structure of the polyhydroxylated peptide was studied by spectroscopic methods.

AbstractStarting from the chiral pyrrolidin‐2‐one 2, the carboxy group at C‐4 underwent homologation, and subsequent removal of the 1‐(4‐methoxyphenyl)ethyl group gave lactam 6. Alkylation of N‐1 with benzyl bromoacetate led to 7, a new conformationally restricted analogue of the dipeptide EG (Glu‐Gly).

The effect of an original dimeric dipeptide NGF mimetic, GK-2 (hexamethylenediamide bis-N-monosuccinyl-L-glutamyl-L-lysine, designed on the basis of the beta-turn of the 4th NGF loop) on the hemostasis and fibrinolysis was studied in intact and diabetic Wistar rats in various periods of disorder development.
