The connEulor: Unbiased estimation of connectivity using physical disectors under projection
Bone, 1993The Euler number and the connectivity of an arbitrary object is defined, and it is illustrated why the connectivity of an n-dimensional object cannot be estimated in an (n-1)-dimensional section. The disector--principle for 3-D counting of the Euler--events is illustrated in cancellous bone.

Verification of the disector method for counting neurons, with comments on the empirical method
The Anatomical Record, 1991AbstractThe empirical and disector methods are unbiased sampling methods for determining numbers of neurons. The present study verifies and thus calibrates these methods by determining true numbers of ganglion cells in serial reconstructions and then using each method to estimate the same populations.

Using a stereological method, the optical disector, we examined three inbred strains of mice (NZB/BINJ, DBA/2, and C57BL/6J) for morphological differences in volume, neuronal number, and density of the pyramidal cell and dentate gyrus granule cell layers of the hippocampus. We found significant differences in volume and neuronal number for both regions

Stereological estimation of nucleolar organizer regions in prostatic tissue with an optical disector
APMIS, 1992Silver‐stained nucleolar organizer regions (AgNORs) were studied in 10 hyperplastic, 10 inflamed, nine prostatic intraepithelial neoplastic (PIN) and 20 malignant prostatic lesions. Optical disector measurement on 15 urn sections showed that upper frequency limits for 1.5 um and 2.0 μm AgNOR particles were 10% and 2% in benign lesions and 18% and 9% in

AbstractWe describe MS‐DOS software for the optical volume fractionator (OVF), a stereological method combining the principles of the optical disector (Gundersen et al.: Acta Pathol. Microbiol. Immunol. Scand., 96:857–881, 1988) and fractionator (Gundersen: J. Microsc., 143:3–45, 1986).

Quantification of dendritic spine populations using image analysis and a tilting disector
Journal of Neuroscience Methods, 1995A series of image analysis routines, stochastic geometry methodology, and a design-based stereological procedure have been developed to quantify objectively the length, layout, and the true density of neuronal dendritic spines observed at the light (or confocal) microscope level.

Design-based stereology is considered the gold-standard method for estimating the total number of glomeruli, and thereby nephrons, in the adult kidney. However, until recently, a design-based method for estimating nephron number in the developing kidney was not available.

Efficient estimation of number density in opaque material microstructures: the large‐area disector
Journal of Microscopy, 2000Unbiased and efficient estimation of number density of features in opaque material microstructures has been quite difficult. In this contribution a montage‐based efficient serial sectioning technique is presented as a practical solution for efficient estimation of number density in such microstructures.

A quantitative description of a three-dimensional (3D) object based on two-dimensional images can be made using stereological methods These methods involve unbiased approaches and provide reliable results with quantitative data. The quantitative morphology of the nervous system has been thoroughly researched in this context.

The total number of neurons in the human neocortex unbiasedly estimated using optical disectors
Journal of Microscopy, 1990SUMMARYAn efficient method is presented for obtaining, in under 4h, an unbiased estimate of the total number of neurons in the human neocortex, with a coefficient of error on the estimate of ∼ 5%. The novel sampling scheme used in this study is unbiased and was designed so that only a small amount of neocortical grey matter had to be removed.
