ATP-type DNA ligase requires other proteins for its activity in vitro and its operon components for radiation resistance in Deinococcus radiodurans in vivo

Biochemistry and Cell Biology, 2010
A multiprotein DNA processing complex isolated from Deinococcus radiodurans contains the DNA repair protein PprA, an ATP-type DNA repair ligase (LigB) encoded by the drB0100 gene, and protein kinase activity. An ATP-dependent DNA end-joining activity was detected in the complex.

A sensitive ligase-based ATP electrochemical assay using molecular beacon-like DNA

open access: closedBiosensors and Bioelectronics, 2010
A sensitive and selective ligase-based signal-on electrochemical sensing method for adenosine-5'-triphosphate (ATP) detection had been developed using molecular beacon (MB)-like DNA. In this method, the biotin-tagged MB-like DNA was self-assembled onto a gold electrode to form a stem-loop structure by means of facile gold-thiol chemistry, which ...

Interaction of β-L-Adenosine-5′-triphosphate (L-ATP) with Human Deoxycytidine Kinase, Human DNA Primase and T4 DNA Ligase: Does the Chance Direct Enzymatic Enantioselectivity?

Nucleosides and Nucleotides, 1999
We demonstrate that L-ATP: 1) as well as its natural D-enantiomer, acts as a phosphate donor in the reaction catalysed by human deoxycytidine kinase; 2) inhibits human DNA-primase and the ATP-dependent T4 DNA ligase. Thus, the lack of enantioselectivity of the enzymes is more frequent than it was believed a few years ago and we suggest that it would ...

Functional domains of an ATP-dependent DNA ligase.

Journal of molecular biology, 1999
The crystal structure of an ATP-dependent DNA ligase from bacteriophage T7 revealed that the protein comprised two structural domains. In order to investigate the biochemical activities of these domains, we have overexpressed them separately and purified them to homogeneity. The larger N-terminal domain retains adenylation and ligase activities, though

Synthesis and bioevaluation of aryl hydroxamates distinguishing between NAD+ and ATP-dependent DNA ligases

MedChemComm, 2012
Identification of compounds distinguishing between NAD+ and ATP-dependent ligases is a key factor to discover new antimicrobials. Based on virtual screening experiments a series of hydroxamates distinguishing between the above two classes of enzymes are synthesized.

A new group of plant-specific ATP-dependent DNA ligases identified by protein phylogeny, hydrophobic cluster analysis and 3-dimensional modelling

Functional Plant Biology, 2005
The eukaryotic ATP-dependent DNA ligases comprise a group of orthologous proteins that have distinct roles in DNA metabolism. In contrast with the well-known DNA ligases of animal cells, the DNA ligases of plant cells are poorly described. Until now, only two DNA ligases (I and IV) genes of Arabidopsis thaliana (L.) Heynh were isolated and ...

Rapid Time Scale Analysis of T4 DNA Ligase-DNA Binding.

Biochemistry, 2017
DNA ligases, essential to both in vivo genome integrity and in vitro molecular biology, catalyze phosphodiester bond formation between adjacent 3'-OH and 5'-phosphorylated termini in dsDNA.
