Hepatocellular carcinoma (HCC)‐derived extracellular vesicles (EVs) enrich the metabolic enzyme ATP‐citrate lyase (ACLY). EV‐transferred ACLY enhances palmitate biosynthesis, increases the S‐palmitoylation and stability of multiple immune checkpoint proteins, augments the cellular immunosuppressive activity, and ultimately accelerates the malignant ...

First Generation Proteolysis Targeting Chimeras (PROTACs) for the Treatment of Progeria
We report the first PROTACs designed to degrade progerin, introducing a novel therapeutic approach for progeria. The best compound, UCM‐18142, significantly reduces progerin levels and improves key disease phenotypes in patient‐derived cells and in the LmnaG609G/G609G mouse model, paving the way for new treatment strategies targeting the root cause of ...

DNA Replication Errors Drive Genome‐Wide Small Inverted Triplication Dynamics
This study provides insight into the dynamic equilibrium mechanism of a novel structural variant, small inverted triplication (SIT), which is generated by misalignment of the 3’ flap generated under DNA replication stress with palindromic sequence. Alternatively, the end sequence may fold back on itself to form an inverted fragment.

Producing MSCs on rigid culture substrates induces a scar‐making phenotype, jeapordizing therapeutic success. ‘Tissue‐soft’ surfaces prevent MSC fibrogenesis and preserve regenerative traits. An epigenetic network, driven by HOXA11 and SALL1, maintains ‘soft memory’ by keeping chromatin open in relaxed MSCs, promoting anti‐fibrotic programs.

Multidrug‐resistant Vibrio infections are rising rapidly and threaten coastal populations worldwide. This study introduces D‐zp37, a chirality‐engineered antimicrobial peptide with exceptional potency against resistant Vibrio species. D‐zp37 kills planktonic cells, blocks mixed‐species biofilms, disrupts essential bacterial stress responses, and shows ...

We report a rational design strategy for STING antagonists by dual covalent targeting of Cys292/309 in its C‐terminal domain, directly preventing functional oligomerization. Through covalent warhead repurposing, we identified P005091 and revealed its unique dual‐cysteine mechanism.

In colorectal cancer, high expression of the DNA repair protein EEPD1 correlates with immune exclusion and poor prognosis. This study demonstrates that EEPD1 depletion induces genomic instability, leading to cytosolic DNA accumulation and subsequent activation of the cGAS‐STING‐type I interferon pathway. This cascade remodels the tumor microenvironment

Strand displacement reactions are important bricks for the construction of various DNA nanodevices, among which the toehold-mediated strand displacement reaction is the most prevalently adopted. However, only a limited number of tools could be used to finely regulate the toehold reaction, thus restricting DNA nanodevices from being more multifunctional

The neuronal perceptron with DNA strand displacement
2018 Tenth International Conference on Advanced Computational Intelligence (ICACI), 2018In the field of biologic computing and neural networks, DNA strand displacement technology performs well in computing, programming and information processing. In this paper, based on two-domain DNA strand displacement, the neural DNA perceptron is proposed to implement Boolean operations. Some operation instances are simulated by Visual DSD (DNA strand
