Direct monitoring of the strand passage reaction of DNA topoisomerase II triggers checkpoint activation. [PDF]
By necessity, the ancient activity of type II topoisomerases co-evolved with the double-helical structure of DNA, at least in organisms with circular genomes.

A synthetic benzoxazine dimer derivative targets c‐Myc to inhibit colorectal cancer progression
Benzoxazine dimer derivatives bind to the bHLH‐LZ region of c‐Myc, disrupting c‐Myc/MAX complexes, which are evaluated from SAR analysis. This increases ubiquitination and reduces cellular c‐Myc. Impairing DNA repair mechanisms is shown through proteomic analysis.

DNA topoisomerase II catalyzes two different chemical reactions as part of its DNA transport cycle: ATP hydrolysis and DNA breakage/religation. The coordination between these reactions was studied using mutants of yeast topoisomerase II that are unable ...

Mechanism of type II DNA topoisomerases: a tale of two gates [PDF]
DNA topoisomerases are fascinating enzymes involved in many cellular processes including replication, transcription, and genetic recombination. They are capable of catalyzing topological rearrangements of DNA such as relaxation of supercoiled DNA and catenation and decatenation of DNA rings [1xCozzarelli, N.R. and Wang, J.C. (eds). Etoposide induces DNA damage, activating p53‐dependent apoptosis via caspase‐3/7, which cleaves PARP1. Dammarenediol II enhances this apoptotic pathway by suppressing O‐GlcNAc transferase activity, further decreasing O‐GlcNAcylation. The reduction in O‐GlcNAc levels boosts p53‐driven apoptosis and influences the Akt/GSK3β/mTOR signaling pathway ...

A nucleotide resolution map of Top2-linked DNA breaks in the yeast and human genome [PDF]
DNA topoisomerases are required to resolve DNA topological stress. Despite this essential role, abortive topoisomerase activity generates aberrant protein-linked DNA breaks, jeopardising genome stability.

Chemotherapies such as doxorubicin can have toxic effects on healthy cardiovascular/heart tissue. Following up on a doxorubicin toxicity study in mice without tumors where nitrate water was cardioprotective (lessened toxicity), this study with tumor‐bearing mice undergoing doxorubicin treatment showed no negative effect of nitrate and nitrite on drug ...

A 3D biomimetic OS model was developed by bioprinting an OS‐cell‐derived dECM‐enriched bioink with OS spheroids incorporated. The model showed upregulation of known OS prognostic markers and increased resistance to doxorubicin, compared to 2D cultures and scaffold‐free spheroids, making this a more clinically relevant platform for drug discovery ...
