ABSTRACT Background While Wilms tumor (WT) typically has a favorable prognosis, relapsed cases—especially those with high‐risk histology—remain therapeutically challenging after intensive frontline therapy. The combination of vincristine and irinotecan has demonstrated activity in pediatric solid tumors, and pazopanib, a multi‐targeted tyrosine kinase ...

Drug tolerance is an acute defence response preceding a fully drug-resistant state and tumour relapse; however, there are few therapeutic agents targeting drug tolerance in the clinic.

ABSTRACT Objectives To identify predictors of chronic ITP (cITP) and to develop a model based on several machine learning (ML) methods to estimate the individual risk of chronicity at the timepoint of diagnosis. Methods We analyzed a longitudinal cohort of 944 children enrolled in the Intercontinental Cooperative immune thrombocytopenia (ITP) Study ...

ABSTRACT Introduction Neuroblastoma (NB) with central nervous system (CNS) metastases is rare at diagnosis, but occurs more often during relapse/progression. Patients with CNS metastases face a dismal prognosis, with no standardized curative treatment available.

Drug Tolerance in Mycobacterium tuberculosis [PDF]
ABSTRACT Although Mycobacterium tuberculosis is eradicated rapidly during therapy in some patients with pulmonary tuberculosis, it can persist for many months in others. This study examined the relationship between mycobacterial drug tolerance (delayed killing in vitro), persistence, and relapse.

Pathogenic Germline PALB2 and RAD50 Variants in Patients With Relapsed Ewing Sarcoma
ABSTRACT Approximately 10% of patients with Ewing sarcoma (EwS) have pathogenic germline variants. Here, we report two cases: first, a novel germline pathogenic variant in partner and localizer of BRCA2 (PALB2) in a patient with a late EwS relapse. Its impact on homologous recombination is demonstrated, and breast cancer risk is discussed.

Methadone tolerance testing in drug misusers [PDF]
Asking drug misusers the dose they need for methadone maintenance might seem like an invitation to exaggerate, but experience in one practice suggests that it could improve uptake of treatment without compromising ...

Targeting redox heterogeneity to counteract drug tolerance in replicating Mycobacterium tuberculosis
Chloroquine resets the redox physiology of Mycobacterium tuberculosis to potentiate the efficacy of antibiotics in vivo. Antibiotic redox-based redux Phagosomal pH and redox heterogeneity in Mycobacterium tuberculosis (Mtb) can promote tolerance of the ...

KDM5A and KDM5B histone-demethylases are overexpressed in many cancers and have been involved in drug tolerance. Here, we describe that KDM5A, together with KDM5B, contribute to replication stress (RS) response and tolerance.
